      Potensi Hidrolisat Protein Maggot (Hermetia illucens) Sebagai Antihiperglikemia

      Abstract
      Penelitian ini bertujuan mendapatkan hidrolisat protein maggot terbaik dan menguji kemampuan efek hipoglikemik serta menentukan dosis optimumnya. Hidrolisat protein maggot dibuat menggunakan reaksi hidrolisis enzimatis tripsin selama 3, 5 dan 7 jam. Kemudian hasil ke 3 jenis hidrolisat protein maggot diidentifikasi menggunakan LC-MS/MS dan dibandingkan jumlah asam amino insulinotropiknya untuk memperoleh satu jenis sampel terbaik. Hidrolisat terbaik adalah yang dihidrolisis selama 7 jam karena mengandung 6 jenis asam amino insulinotropik tertinggi dari pada ke 2 sampel lainnya, sehingga dapat dilanjutkan ke uji efek hipoglikemik pada tikus hiperglikemia. Pengujian tersebut menggunakan metode oral glucose tolerance test. Sejumlah 21 ekor tikus jantan galur Sprague dawley telah digunakan pada penelitian ini, yang dikelompokkan menjadi tujuh kelompok perlakuan. Kelompok perlakuan meliputi kontrol positif, kontrol negatif, kontrol obat akarbose dan 4 kelompok perlakuan hidroliat protein, mulai dari dosis 100, 200, 300, dan 400 mg/kg berat badan. Penelitian ini menggunakan rancangan acak lengkap yang selanjutnya dianalisis dengan ANOVA dan uji beda Duncan jika terdapat pengaruh perlakuan. Pemberian hidrolisat protein maggot dengan dosis 100, 200, 300, dan 400 mg/kg bobot badan tikus menunjukkan penurunan kadar glukosa darah. Dosis 300 mg/kg berat badan merupakan dosis terbaik karena menunjukkan pengaruh hipoglikemik optimum pada tikus hiperglikemia.
       
      This study aims to obtain the best maggot protein hydrolyzate and test the ability of the hypoglycemic effect and determine the optimum dose. Maggot protein hydrolyzate was prepared using trypsin enzymatic hydrolysis reaction for 3, 5 and 7 hours. Then the results of the 3 types of hydrolyzate were identified and compared using LC-MS/MS. The best hydrolyzate is the one that is hydrolyzed for 7 hours because it contains the highest 6 types of insulinotropic amino acids, so that it can be continued to test the hypoglycemic effect in hyperglycemic rats. The test uses the oral glucose tolerance test method. A total of 21 male Sprague dawley rats were used in this study, which were divided into seven treatment group. Each treatment group had 3 replications. This study used a completely randomized design which was further analyzed by ANOVA and Duncan's difference test, if there was an effect of treatment. The administration of maggot protein hydrolyzate at doses of 100, 200, 300, and 400 mg/kg body weight of the rats showed a decrease in blood glucose levels. The dose of 300 mg/kg body weight is the best dose because it shows the optimum hypoglycemic effect in hyperglycemic rats.
