      Kesesuaian Dua Kit ELISA Komersial untuk Deteksi Antibodi terhadap Virus Zika

      Abstract
      Virus Zika (ZIKV) merupakan salah satu virus dari keluarga Flaviviridae yang dapat ditularkan melalui gigitan nyamuk. Antibodi ZIKV tidak hanya ditemukan pada sampel serum manusia tetapi ditemukan juga pada serum satwa primata. Salah satu metode uji serologi untuk mendeteksi dan mengukur titer antibodi ZIKV dilakukan dengan Enzyme-Linked Immunosorbent Assay (ELISA). Kit ELISA komersial untuk ZIKV telah banyak beredar di pasaran. Dalam pengujian ELISA diperlukan kit yang berkualitas baik sehingga mampu memberikan hasil uji yang akurat. Penelitian ini bertujuan untuk menentukan kesesuaian antara dua kit ELISA ZIKV komersial untuk melihat apakah dalam penggunaannya dapat saling menggantikan. Kit ELISA yang digunakan yaitu: kit A dan B; yang diuji dengan 83 sampel serum satwa primata yang sama. Kesesuaian ditentukan berdasarkan perhitungan nilai Cohen’s Kappa dengan menggunakan Microsoft Exel 2019. Hasil uji Kappa menunjukkan bahwa kit A dan B menghasilkan nilai 0.368 (fair agreement). Nilai Kappa yang diperoleh kedua kit tersebut menghasilkan nilai kesesuaian yang rendah, menunjukkan bahwa kedua kit tersebut tidak dapat saling menggantikan.
       
      Zika virus (ZIKV) is a species of Flaviviridae family commonly transmitted by mosquito bites. ZIKV antibody not only can be detected in human serum samples but also nonhuman primates. One of the methods to detect and measure ZIKV antibody through the serology test is the Enzyme-Linked Immunosorbent Assay (ELISA). There are many commercial ELISA kits for ZIKV in the market. A good quality kit is needed to provide accurate test results. This study was aimed to determine the compatibility of two commercial ZIKV ELISA kits to see if they can be used interchangeably. ZIKV ELISA kits were used, namely: Kits A and B; utilizing the same 83 samples of nonhuman primate serum tested with the two kits. The compatibility or results agreement of the two kits was determined based on the Cohen's Kappa value through calculation with Microsoft Excel 2019. Kappa test showed that kits A and B resulted a value of 0.368 which fell into criteria as fair agreement. The Kappa value obtained by the two kits resulted in a low suitability value, suggesting that the two kits cannot be used interchangeably.
