dc.description.abstractPerubahan organ yang terjadi setelah kematian dapat memperkirakan interval waktu kematian. Penelitian ini bertujuan mengamati perubahan histopatologi yang terjadi pada organ ginjal sampai 48 jam setelah kematian / post mortem (PM). Sebanyak 75 ekor ayam pedaging umur 7 hari secara bersamaan dieuthanasi dengan cara dislokasi persendian leher. Nekropsi dilakukan pada 3 ekor ayam setiap selang waktu 2 jam mulai jam ke-0 sampai jam ke-48. Organ ginjal hasil nekropsi difiksasi dengan cara direndam dalam Buffer Neutral Formaline (BNF) 10%, lalu diproses menjadi preparat histopatologi untuk diamati perubahan pada organ ginjal secara mikroskopis. Hasil pengamatan dianalisis menggunakan aplikasi Image J dan uji korelasi pearson. Kematian sel-sel di organ ginjal setelah kematian menyebabkan pelebaran ruang Bowman serta penurunan kerapatan dan perubahan warna parenkim ginjal. Hasil menunjukkan lisis kapsula Bowman terjadi pada jam ke-18 PM, munculnya bakteri disertai penggelapan warna jaringan pada jam ke-24 PM dan kerapatan parenkim ginjal < 50% terjadi pada jam ke-46 PM. Dapat disimpulkan bahwa terdapat derajat perubahan histopatologi yang berurutan sesuai lamanya waktu kematian ayam sampai 48 jam PM. Perubahan yang terjadi tersebut dapat digunakan sebagai parameter dalam menentukan waktu kematian ayam. Kata kunci: ayam pedaging, ginjal, histopatologi, pembusukan, serial waktuid
dc.description.abstractChanges in organ after death can be used to predict the time of death interval. The aim of the present study is to examine the histopathological lesion of kidney up to 48 hours after death / post mortem (PM). A total of 75 broilers aged 7 days were simultaneously euthanized by neck dislocation. Necropsy was performed on 3 chickens every 2 hours from 0 to 48 hours. The kidneys obtained from the necropsy were fixed in a Buffer Neutral Formaline 10%, and then processed into histopathological preparations to observe the structural changes in kidneys microscopically. The results were analyzed using ImageJ and Pearson correlation test. Cell death in kidney caused dilation of the Bowman space and a decrease in the density and discoloration of the renal parenchyma. The results showed that Bowman’s capsule lysis at 18th hours PM, the appearance of bacteria accompanied with darkening of the tissue observed at 24th hours PM and renal parenchymal density < 50% observed at 46th hours PM. It can be concluded that, there were a series of histopathological changes in kidney within 48 hours PM.
