Banana is the most consumed of horticulture commodity plants. The government and society also cultivate some banana varieties such as Cavendish, Raja Buluh, and Barangan. Considering the production demand is also increased, the government also implemented the fruit self-sufficiency program in the Minister of Plantation Regulation Number 86/Permentan/OT.140/8/2013. Therefore, BUMN began to cultivate banana plants in several plantation sectors owned by the company. During the banana plants’ cultivation process, many problems are faced, one of which is the banana plant attacked by a pathogen. The primary pathogen that attacks the banana plant is Fusarium oxysporum causing the fusarium wilt. F. oxysporum is categorized as the soil-borne pathogen that infected banana plant through the wound of plant tissue. The wound found in the plants’ root could be identified as the natural wound result of plant growth process or caused by several vectors such as nematode. Many researchers reported that nematode has a significant role in the process of fusarium wilt disease, which is caused by F. oxysporum. This research aims to observe the abundance of F. oxysporum and phytonematode founded in the banana plant and the relation of both pathogens in causing fusarium wilt. The hypothesis of this research that phytonematode founded a positive correlation toward the severity of fusarium wilt disease.
The method used in this research was observing the severity of fusarium wilt of banana plant Emas Kirana varieties that’s most know as Mas Kirana. Based on the predetermined score rating. Soil and root samples was collected which was conducted by purposive method. From every block, samples were taken from 5 different clumps with different disease severities. Samples were collected from 5 different blocks as replication. Sample from 5 different blocks with the same disease severity were then mix for future research. The abundance of F. oxysporum and phytonematodes were calculated by applying conventional and molecular methods. Isolation of phytonematodes from soil were conducted by extracting the phytonematodes from soil samples by using a floatation centrifugation method. Identification of phytonematodes to the genus level were carried out by observing the morphology of their anterior to posterior. The abundance of phytonematodes then calculated under microscope by placed 1 ml of phytonematode suspension on cyrus dish, and repeated 3 times. Furthermore, the number of phytonematodes was calculated using the formula of absolute population calculation. Calculation of F. oxysporum was conducted by two ways, conventional (growing on synthetic media) and molecular methods. The conventional method was performed by serial dilution of soil samples, followed by cultured on Martin Agar, and incubated for 3-4 days before counting the number of F. oxysporum colonies grow on medium. To prove that F. oxysporum growing on Martin Agar is Foc, molecular identification was carried out using specific primer Foc1/F (5’-CAGGGGATGTATGAGGAGGCT-3’) and Foc2/R (3’-GTGACAGCGTCGTCTAGTTCC-5’). PCR result then visualized on agarose gels.
Real-time PCR technique was also conducted to count the abundance of F. oxysporum from soil samples. The standard curve construction is started by isolating the competent bacteria cell of DH5 alpha that has been added the synthesis gene of F. oxysporum. The cell transformation is becoming plasmid that has F. oxysporum gene and isolation of DNA plasmid using lysis alkaline method. DNA plasmid is made by diluting 10-4. Estimation of number of F. oxysporum obtained from copies number plasmid formula. Statistical analysis was performed by using regression and correlation methods in the Microsoft Excel and PLS-SEM application to examine the relationship between abundance of F. oxysporum and phytonematodes against fusarium wilt disease severity scoring.
Phytonematodes obtained in this research are Helicotylenchus sp., Criconemoides sp. and Radopholus sp. In the soil, the highest phytonematodes population was found at plant with disease severity score of 4 as many as 77 individuals per g of soil. The lowest population was found at plants with score of 2 as many as 16 individuals per g of soil. At the plant roots, the highest phytonematodes population was at plants with score of 0 as many as 85 individuals per g of root. And the lowest population was at score of 3 as many as 33 individuals per g of root. Based on colonies characteristics and microscopic morphologies, it was concluded that fungus grows on Martin Agar is F. oxysporum. Application of pairs of specific primer also proved that it is F. oxysporum strain 4 (tropical strain 4). Calculation of F. oxysporum using pouring plate methods on Martin Agar found that the highest population of F. oxysporum was at plants with disease severity score of 1 as much as 8.1x103 cfu per g of soil and the lowest at score of 0 as much as 1.0x 103 cfu per g of soil. Calculation with real-time PCR found that the highest population of F. oxysporum was at plants with disease severity score of 0 as much as 2.663x1014 ng per ul and the lowest population at score of 4 as much as 6.06x1013 ng per ul. The result of calculating the abundance of F. oxysporum on both methods could not be compared because of the differences in the measured portion, pour plate method counted the living fungal propagules, while real-time PCR method calculates the portion of DNA of F. oxysporum amplified by primer used.
There was no correlation between the presence of F. oxysporum and phytonematodes in the soil in an increase of disease severity of fusarium wilt of banana. After being analyzed by PLS-SEM, both the counted pathogen population were negatively correlated, with the value of -1.5%. F. oxysporum could attack the banana plant and causes the fusarium wilt without help of phytonematode with the value of 8.3%. The presence of phytonematodes in the soil has no role to the occurrence of fusarium wilt of banana at PTPN VIII Parakansalak plantation. The abundance of phytonematodes and F. oxysporum in the soil and plant tissues could not describe the level of disease severity of fusarium wilt of banana. Pisang termasuk komoditi tanaman hortikultura yang banyak dikonsumsi masyarakat sehingga permintaan produksi pisang menjadi meningkat. Pemerintah maupun masyarakat di Indonesia membudidayakan beberapa varietas pisang seperti Cavendish, Raja Buluh, dan Barangan. Selain permintaan produksi yang meningkat, langkah ini dilakukan pemerintah untuk melakukan swasembada buah yang tertera pada Peraturan Menteri Pertanian Nomor 86/Permentan/OT.140/8/2013. Oleh karena itu, pihak BUMN mulai melakukan budidaya tanaman pisang pada beberapa sektor kebun yang dimiliki oleh perusahaan. Selama proses budidaya banyak permasalahan yang dihadapi, patogen utama yang menyerang tanaman pisang salah satunya Fusarium oxysporum. Patogen ini yang menyebabkan penyakit layu fusarium. Cendawan F. oxysporum termasuk patogen tular tanah yang menginfeksi tanaman pisang melalui luka pada jaringan tanaman. Luka pada perakaran tanaman dapat berupa luka alami karena proses pertumbuhan tanaman atau disebabkan oleh beberapa vektor seperti nematoda. Beberapa penelitian menyatakan bahwa nematoda memiliki peran penting dalam proses infeksi penyakit layu fusarium oleh F. oxysporum. Tujuan dari penelitian ini adalah mengamati kelimpahan F. oxysporum dan fitonematoda pada pertanaman pisang dan hubungan kedua patogen dalam menyebabkan layu fusarium. Hipotesis dari penelitian ini populasi fitonematoda pada pertanaman pisang berkolerasi positif terhadap keparahan penyakit layu fusarium yang terjadi di lapangan. Metode yang dilakukan pada penelitian ini adalah mengamati tanaman pisang pada varietas Emas Kirana yang dikenal dengan Mas Kirana. Pengamatan berdasarkan skoring keparahan penyakit layu fusarium yang telah ditentukan sebelumnya. Pengambilan sampel pada bagian tanah dan akar tanaman pisang dilakukan secara purposive sampling. Pada tiap blok diambil sampel dari 5 rumpun berbeda dengan keparahan penyakit yang berbeda. Sampel dikumpulkan dari 5 blok berbeda sebagai ulangan. Sampel dari 5 blok berbeda dengan keparahan penyakit yang sama lalu dikompositkan untuk penelitian selanjutnya. Penghitungan kelimpahan F. oxysporum dan fitonematoda menggunakan metode konvensional dan molekuler. Isolasi fitonematoda dari tanah dilakukan dengan ekstraksi fitonematoda menggunakan metode flotasi sentrifugasi. Identifikasi fitonematoda sampai tingkat genus dengan mengamati morfologi secara berurut dari anterior ke posterior. Analisis kelimpahan fitonematoda dilakukan dengan menghitung 1 ml suspensi fitonematoda yang diletakkan pada cawan sirakus dan diulang sebanyak 3 kali. Selanjutnya, jumlah fitonematoda dihitung menggunakan rumus penghitungan populasi absolut. Isolasi F. oxysporum dilakukan menggunakan 2 metode yaitu secara konvensional dan molekuler. Metode konvensional cendawan F. oxysporum ditumbuhkan pada media agar Martin dan diinkubasi selama 3-4 hari. Identifikasi dan perhitungan kelimpahan F. oxysporum dilakukan pada isolat yang memenuhi karakter dengan diberi tanda. Secara molekuler, ekstraksi DNA dari sampel tanah yang terinfeksi layu fusarium, amplifikasi PCR menggunakan primer spesifik dengan urutan basa Foc1/F (5’-CAGGGGATGTATGAGGAGGCT-3’) dan Foc2/R (3’-GTGACAGCGTCGTCTAGTTCC-5’) dan visualisasi hasil PCR. Kelimpahan F. oxysporum juga dihitung menggunakan teknik real-time PCR. Konstruksi kurva standar diawali dengan isolasi kompeten sel bakteri DH5 alpha yang telah ditambahkan gen sintesis F. oxysporum. Selanjutnya transformasi sel menjadi plasmid yang telah mengandung gen F. oxysporum dan isolasi DNA plasmid menggunakan metode lisis alkalin. DNA plasmid diencerkan hingga pengenceran 10-4. Estimasi jumlah plasmid F. oxysporum yang diperoleh dihitung menggunakan rumus copies number plasmid. Analisis statistik dilakukan menggunakan metode regresi dan korelasi pada Microsoft Excel dan aplikasi PLS-SEM untuk melihat hubungan antara kelimpahan F. oxysporum dan fitonematoda terhadap skoring keparahan penyakit layu fusarium. Fitonematoda yang diperoleh pada penelitian ini yaitu Helicotylenchus sp., Criconemoides sp. dan Radopholus sp. Pada tanah populasi fitonematoda tertinggi pada tanaman skor 4 sebanyak 77 individu per g tanah dan yang terendah pada skor 2 sebanyak 16 individu per g tanah. Pada akar populasi fitonematoda tertinggi pada tanaman skor 0 sebanyak 85 individu per g tanah dan yang terendah pada skor 3 sebanyak 33 individu per g tanah. Pada penghitungan dengan metode cawan tuang, ditemukan bahwa populasi paling tinggi pada tanaman skor 1 sebesar 8.1x103 cfu per g tanah dan terendah pada tanaman skor 0 dengan nilai 1.0x103 cfu per g tanah. Sedangkan secara molekuler, hasil identifikasi menunjukkan patogen termasuk F. oxysporum TR4 dan populasinya paling tinggi pada skor 0 sebesar 2.663x1014 ng per ul dan terendah pada skor 4 sebesar 6.06x1013 ng per ul. Hasil penghitungan kelimpahan F. oxysporum pada kedua metode yang digunakan tidak dapat dibandingkan karena perbedaan bagian yang diukur, metode cawan tuang menghitung propagul cendawan yang masih hidup, sedangkan metode real- time PCR menghitung bagian DNA F. oxysporum yang teramplifikasi dengan primer yang digunakan. Tidak ditemukan adanya korelasi antara keberadaan F. oxysporum dan fitonematoda di dalam tanah dalam peningkatan keparahan penyakit layu fusarium pada pisang. Setelah dianalisis dengan PLS-SEM, kedua populasi patogen yang dihitung berhubungan negatif, dengan nilai sebesar –1.5%. Cendawan F. oxysporum dapat menyerang tanaman pisang dan menyebabkan penyakit layu fusarium tanpa bantuan fitonematoda dengan nilai sebesar 8.3%. Fitonematoda tidak berperan dalam terjadinya penyakit layu fusarium pada pisang di perkebunan PTPN VIII Parakansalak. Kepadatan fitonematoda dan F. oxysporum dalam tanah dan jaringan tanaman pisang tidak dapat menggambarkan tingkat keparahan penyakit layu fusarium pada pisang.
