      Minyak Kelapa Kopyor sebagai Substrat Baru untuk Biosintesis Flavor dari Yarrowia lipolytica dalam Teknologi Pangan

      Abstract
      Yarrowia lipolytica dapat tumbuh pada berbagai substrat organik, seperti minyak nabati, lemak hewani, dan senyawa hidrokarbon. Media Yeast Nitrogen Base without Amino Acid (YNB) digunakan untuk mendukung metabolisme khamir dan meningkatkan produksi bioflavor dalam proses fermentasi. Media tersebut memiliki harga yang relatif tinggi, sehingga dibutuhkan alternatif media pertumbuhan yang lebih terjangkau. Berbagai substrat telah diteliti untuk mengetahui potensi substrat alternatif sebagai media pertumbuhan khamir. Pada penelitian ini, pemanfaatan substrat minyak kelapa kopyor digunakan sebagai alternatif media pertumbuhan. Tujuan penelitian ini adalah meneliti penggunaan media substitusi minyak kelapa kopyor sebagai substrat baru untuk biosintesis flavor dari Yarrowia lipolytica melalui pengamatan tingkat pertumbuhan khamir, berat basah sel, kerapatan sel, morfologi mikroskopis, dan menganalisis senyawa flavor yang terbentuk selama proses fermentasi. Perlakuan pada penelitian ini menggunakan tiga taraf konsentrasi, dua varietas kelapa kopyor, dan variasi waktu fermentasi. Hasil penelitian menunjukkan substitusi minyak kelapa kopyor dengan konsentrasi 7,5% memberikan peningkatan pertumbuhan Yarrowia lipolytica yang lebih tinggi dibandingkan media kontrol. Minyak kelapa kopyor dari varietas Dalam menghasilkan pertumbuhan yang lebih tinggi dibandingkan dengan varietas Genjah pada semua konsentrasi. Hasil identifikasi senyawa hasil fermentasi mengungkapkan bahwa teridentifikasi berbagai senyawa bioflavor, seperti asam fumarat, propanoat, karbonat, gliserol trikaprilat, senyawa fenol, dan senyawa- senyawa lainnya.
       
      Yarrowia lipolytica can grow on various organic substrates, such as vegetable oils, animal fats, and hydrocarbon compounds. Yeast Nitrogen Base without Amino Acids (YNB) is used to support yeast metabolism and enhance bioflavor production in the fermentation process. However, this medium is relatively expensive, thus necessitating more affordable alternative growth media. Various substrates have been studied to explore their potential as alternative growth media for yeast. Therefore, utilizing kopyor coconut oil as a substrate could serve as an alternative growth medium. The purpose of this study was to examine the use of kopyor coconut oil substitution media as a new substrate for flavor biosynthesis from Yarrowia lipolytica by observing yeast growth rate, wet cell weight, cell density, microscopic morphology, and analyzing the flavor compounds formed during the fermentation process. The experiment involved three concentration levels, two kopyor coconut varieties, and variations in fermentation time. The results showed that kopyor coconut oil substitution at a 7.5% concentration significantly increased the growth of Yarrowia lipolytica compared to the control medium. Kopyor coconut oil from the Dalam variety resulted in higher growth compared to the Genjah variety at all concentrations. The identification of fermentation products revealed the presence of various bioflavor compounds, such as fumaric acid, propanoate, carbonate, glycerol tricaprylate, phenolic compounds, and other compounds.
