Pengaruh Kekasaran Permukaan Stainless Steel dan Matriks Pangan terhadap Inaktivasi Biofilm Bacillus cereus Oleh Sanitizer Asam Perasetat
Abstract
Bacillus cereus adalah bakteri patogen penyebab penyakit bawaan pangan
karena kemampuannya memproduksi toksin emetik dan enterotoksin. Pada industri
susu, B. cereus telah dilaporkan dapat membentuk biofilm pada stainless steel (SS)
meskipun SS dianggap sebagai bahan peralatan yang paling baik dan sering
digunakan. Keberadaan goresan dan zat gizi dari matriks pangan pada permukaan
diketahui dapat meningkatkan pembentukan biofilm sehingga memerlukan cara
pembersihan yang tepat. Asam perasetat dilaporkan dapat menurunkan jumlah
biofilm B. cereus, namun belum diketahui efektivitasnya dengan pengaruh
permukaan kasar dan matriks pangan. Penelitian ini bertujuan untuk mengetahui
pengaruh kekasaran permukaan dan matriks pangan terhadap pembentukan biofilm
B. cereus pada permukaan stainless steel, konsentrasi dan waktu kontak asam
perasetat untuk menginaktivasi biofilm B. cereus pada stainless steel, serta
pengaruh kekasaran permukaan stainlees steel dan matriks pangan terhadap
efektivitas inaktivasi biofilm B. cereus oleh asam perasetat. Penelitian ini
menggunakan B. cereus LH-10KP1 yang diisolasi dari susu pasteurisasi dan asam
perasetat komersial. Penelitian terdiri dari tiga tahapan yakni persiapan kultur,
pembentukan biofilm pada permukaan SS halus maupun kasar, dan pengaruh
kekasaran SS dan matriks pangan terhadap inaktivasi biofilm B. cereus dengan
asam perasetat. Pembentukan biofilm dilakukan dengan menginokulasi medium
TSB sehinggu jumlah awal 105 CFU/mL dengan pelat SS halus (normal), kasar,
serta SS halus dengan keberadaan matriks pangan serta menginkubasinya selama
24, 72, 120, serta 168 jam dengan agitasi 70 rpm pada suhu ruang. Inaktivasi biofilm
dilakukan pada biofilm B. cereus berumur 72 jam dengan konsentrasi asam
perasetat 100, 200, dan 400 ppm dan 3 waktu kontak (3, 5, dan 10 menit). Hasil
penelitian menunjukkan densitas biofilm yang terbentuk pada SS halus, kasar, dan
dengan perlakuan matriks pangan berturut-turut 3,4-4,2 log CFU/cm2
, 3,6-4,4 log
CFU/cm2
, serta 3,7-4,2 log CFU/cm2
. Inaktivasi menunjukkan hasil reduksi biofilm
B. cereus meningkat dengan meningkatnya konsentrasi dan waktu kontak asam
perasetat. Reduksi densitas biofilm tertinggi ditemukan pada biofilm yang
dipaparkan pada 400 ppm asam perasetat selama 10 menit yaitu sebesar 3,5 log
CFU/cm2
pada pelat SS halus, serta 3,0 log CFU/cm2 SS kasar dan SS dengan
perlakuan matriks pangan. Peningkatan konsentrasi dan waktu kontak
meningkatkan efektivitas asam perasetat terhadap inaktivasi biofilm B. cereus,
sedangkan perlakuan permukaan kasar dan matriks pangan pada SS menurunkan
efektivitas asam perasetat dalam menginaktivasi biofilm B. cereus. Bacillus cereus is a pathogen that can cause foodborne illness due to its
ability to produce emetic toxins and enterotoxins. In the dairy industry, B. cereus
has been reported to be able to form biofilms on stainless steel (SS) although SS is
considered as the best and most frequently used equipment material. The presence
of scratches and nutrients from the food matrix on the surface is known to increase
the formation of biofilms, thus requiring proper cleaning methods. Peracetic acid is
reported to reduce the amount of B. cereus biofilm, but its effectiveness is not yet
known with the effect of rough surface and food matrix. This study aims to
determine the effect of rough surface and food matrix on B. cereus biofilm
formation on stainless steel, the effect of peracetic acid concentration and contact
time to inactivate B. cereus biofilm on stainless steel, as well as the effect of rough
surface and food matrix on the effectiveness of B. cereus biofilm inactivation using
peracetic acid. This study used B. cereus LH-10KP1 which isolated from
pasteurized milk and commercial peracetic acid. The research involved three stages,
i.e. culture preparation, biofilm formation on smooth and rough SS surfaces, and
the effect of SS roughness and food matrix on the inactivation of B. cereus biofilm
with peracetic acid. Biofilm formation was carried out by inoculating TSB medium
with initial amount of 105 CFU/mL with smooth and rough surfaces, and smooth
SS plates with the presence of the food matrix and incubating it for 24, 72, 120, and
168 hours with 70 rpm agitation at room temperature. Biofilm inactivation was
carried out on B. cereus biofilm aged 72 hours with peracetic acid with the
concentrations of 100, 200, and 400 ppm and 3 contact times (3, 5, and 10 minutes).
The results showed that the density of the biofilm formed on smooth, rough and
food matrix treatment SS surfaces i.e. 3,4-4,2 log CFU/cm2
, 3,6-4,4 log CFU/cm2
,
and 3,7-4,2 log CFU/cm2
. Inactivation showed that the reduction of B. cereus
increased along with increase in peracetic acid concentration and contact time. The
highest reduction of B. cereus biofilm was achieved by PAA at a concentration of
400 ppm for 10 minutes, i.e. 3,5 log CFU/cm2
on smooth SS surface and 3,0 log
CFU/cm2
on rough SS surface and SS plate containing food matrix. This study
suggests that increase on peracetic acid concentration and contact time increase the
effectiveness in inactivating B. cereus biofilm, while rough surface and presence of
food matrix on smooth SS reduce the effectiveness of peracetic acid in inactivating
B. cereus biofilm.
