      Ekstraksi Daun Stevia (Stevia rebaudiana) Menggunakan Metode Enzyme Assisted Extraction (EAE)

      Abstract
      Kebutuhan pemanis gula dari ekstrak tebu terus meningkat setiap tahun, Alternatif pemanis gula adalah pemanis sintesis yang dianggap berbahaya bagi kesehatan apabila dikonsumsi dalam jangka panjang. Stevia rebaudiana memiliki tingkat kemanisan 300 kali gula tebu dan merupakan pemanis alami yang baik untuk kesehatan. Pemanis stevia dapat diperoleh melalui ekstraksi berbantu enzim. Penelitian ini bertujuan untuk mendapatkan pengaruh konsentrasi enzim, waktu pretreatment dan rasio bahan baku-pelarut dalam memperoleh ekstrak steviosida dengan konsentrasi steviosida dan kadar gula total yang tinggi serta kandungan tanin yang rendah. Variabel proses yang digunakan adalah konsentrasi enzim (0.5 dan 1%), waktu pretreatment (10, 20 dan 30 menit), dan rasio bahan-pelarut (1:10, 1:15, dan 1:20 b/v). Ekstraksi daun stevia menggunakan metode EAE terbukti efektif untuk memperoleh konsentrasi steviosida, dikarenakan adanya peningkatan hasil sebesar 3.5 kali lipat dibandingkan dengan ekstraksi tanpa bantuan enzim. Interaksi dari setiap faktor pada kondisi perlakuan (1%;30menit;1:10b/v) memperoleh hasil tertinggi pada konsentrasi steviosida (15.68% b/v) , kadar gula total (5.43% b/v), dan kadar tanin (0.35% b/v).
       
      The need for sugar sweeteners from sugarcane extract continues to increase every year. Alternative sugar sweeteners are synthetic sweeteners that are considered harmful to health if consumed in the long term. Stevia rebaudiana has a sweetness level of 300 times cane sugar and is a natural sweetener that is good for health. Stevia sweeteners can be obtained through enzyme-assisted extraction. This study aims to determine the effect of enzyme concentration, pretreatment time, and raw material-solvent ratio in obtaining stevioside extract with high stevioside concentration and total sugar content, and low tannin content. The process variables used were enzyme concentration (0.5 and 1%), pretreatment time (10, 20 and 30 minutes), and material-solvent ratio (1:10, 1:15, and 1:20 w/v). Extraction of stevia leaves using the EAE method proved to be effective in obtaining stevioside concentrations, due to an increase in yield of 3.5 times compared to extraction without the help of enzymes. The interaction of each factor under treatment conditions (1%;30min;1:10b/v) obtained the highest results at a stevioside concentration of 15.68% (w/v), a total sugar content of 5.43% (w/v), and a tannin content of 0.35% ( b/v).
