Pembuatan Starter Kering dengan Metode Foam-mat Drying untuk Fermentasi Nata De Coco

Proses fermentasi nata de coco pada umumnya menggunakan kultur starter bakteri Acetobacter xylinum dalam bentuk cair. Namun penggunaan kultur starter cair kurang efisien karena memerlukan peremajaan dalam media segar selama waktu tertentu. Metode foam-mat drying atau pengeringan busa digunakan dalam penelitian ini sebagai proses alternatif untuk menghasilkan kultur starter kering. Foam-mat drying adalah proses pembentukan busa stabil dari bahan cair dilanjutkan dengan pengeringan udara. Terdapat dua jenis bahan pengisi yang diujikan yaitu serbuk kelapa kering dan tepung beras. Penelitian ini bertujuan memperoleh formula terbaik dalam proses pembuatan starter kering dengan konsentrasi putih telur yang berbeda untuk menganalisis karakteristik nata de coco yang dihasilkan. Perbandingan bahan pengisi dengan starter cair yang digunakan yaitu 1:2. Kemudian dilakukan pembusaan selama 15 menit dengan penambahan bahan penstabil CMC 0,5%, dan busa putih telur sesuai perlakuan (5%, 10%, 15%, dan 20%). Formulasi dengan bahan pengisi tepung beras steril dan kultur cair 1:2, putih telur 5%, dan inokulasi starter kering sebanyak 5% (b/v) direkomendasikan sebagai formula terbaik, karena menghasilkan busa yang baik serta menghasilkan nata de coco dengan karakteristik fisik terbaik (viabilitas sel 1,8×103 CFU/gram, ketebalan 4,43±0,72 mm, kecerahan 49,44±0,41). The fermentation process of nata de coco generally uses Acetobacter xylinum starter culture in liquid form. Nevertheless liquid starter culture is less efficient because it requires regrowth in fresh media for a certain period. In this study foam-mat drying method was used as an alternative process to produce dried starter. Foam-mat drying is a transformation process of liquid materials to stable foam followed by air drying. There are two types of fillers tested, namely dried coconut powder and rice flour. This study aims to obtain the best formula in the process of dried starter production with different concentrations of egg whites to analyze the characteristics of the nata de coco produced. The ratio of the fillers to the liquid starters used is 1:2. Then continued with the foaming process for 15 minutes with the addition of CMC (0,5%), and egg white according to treatment (5%, 10%, 15%, and 20%). Dried starter formula with rice flour and liquid culture (1:2), 5% egg white, and 5% (w/v) dried starter inoculation is recommended as the best formula, because it produces good foam quality and produces nata de coco with the best physical characteristics (cell viability 1,8×103 CFU/g, thickness 4,43±0,72 mm, and lightness 49,44±0,41).
