Abstract: This research aims to optimize the utilization of sugar cane bagasse through bioconversion into xylooligosaccharides (XOS). XOS production was carried out by enzymatic hydrolysis of sugar cane bagasse with xylanase produced by marine bacteria Bacillus safensis P20. The optimum time to produce xylanase was 24 hours, with xylanase activity of 4.16 U/mL. Xylanase of Bacillus safensis P20 has optimum reaction at pH 7 and 50 °C. The most effective xylanase concentration method was using dialysis with PEG (polyethylene glycol) 6000 due to the highest xylanase activity of 19.62 U/mL and the highest yield of 76.15 %. Hydrolysis process to produce XOS was performed on bagasse substrate concentrations of 0.5 %, 1.0 %, and 1.5 % for 1, 2, and 4 hours. TLC analysis showed that the hydrolysis process was capable to produce oligosaccharides but those were estimated not the type of XOS. The results of HPLC analysis confirmed that the compound formed in hydrolysis process was hydrolyzed xyloglucans (heptasaccharide, octasaccharide, and nonasaccharide).
