Abstract: Urtica dioica L. has long been known as a herbal remedy and nutritious addition to the diet. It has been consumed for a long time as medicinal plants in many parts of the world. This study aimed to observe the effect of U. dioica L. leaf extracts in enhancing proliferation of rat’s spleen lymphocytes. Furthermore, this study also aimed to observe the capability of U. dioica L. leaf extracts in defending lymphocyte against toxic effects of carmoisine and rhodamine B . 30 Sprague Dawley rats were split into three groups, which are control, 0.1 g/kg BW/day dose, 1 g/kg BW/day dose group. The control group was given drinking water while the others were given U. dioica L. leaf crude extracts at their dose respectively. After 90 days of treatment, lymphocyte cells from the rat’s spleen then isolated and tested using MTT proliferation assay with addition of mitogen, and various concentration of carmoisine and rhodamine B. Stimulation index of spontaneous proliferation in 0.1 g/kg BW/day dose group was higher than the other groups. Mitogen-induced proliferation shows an increased proliferation activity in control and 1 g/kg BW/day dose group, while in 0.1 g/kg BW/day dose group a decreased proliferation activity was observed. U. dioica L. leaf crude extracts given at 1 g/kg BW/day shows that the rat’s lymphocyte proliferation activity at the presence of carmoisine was higher than control and 0.1 g/kg BW/day dose group. Likewise, at the presence of rhodamine B, the rat’s lymphocyte proliferation activity of 1 g/kg BW/day dose group was higher than control and 0.1 g/kg BW/day dose group. It is concluded that U. dioica L. leaf crude extract given at 1 g/kg BW/day could inhibit carmoisine and rhodamine B toxic effect on rat’s spleen lymphocytes proliferation activity.
