Abstract: Porphyridium cruentum merupakan mikroalga bersel merah (Rhodophyceae) yang hidup di lingkungan air asin dan menghasilkan polisakarida dan pigmen fikoeritrin. Kultivasi P. cruentum dapat dipengaruhi oleh berbagai faktor, salah satunya cahaya. Lama pencahayaan dapat dimodifikasi selama kultivasi. Tujuan penelitian ini menentukan kurva pertumbuhan P.cruentum, kandungan polisakarida, dan pigmen fikoeritrin berdasarkan lama pencahayaan berbeda. Penelitian ini meliputi 1) kultivasi dan pemanenan P.cruentum dengan perlakuan lama pencahayaan berbeda, yaitu 24:0; 12:12; 20:4 jam terang:gelap ; 2) presipitasi kandungan polisakarida; 3) ekstraksi dan uji konsentrasi pigmen fikoeritrin. Selama pertumbuhan P.cruentum tidak mengalami fase lag. Eksopolisakarida yang dihasilkan adalah 7,93 ± 0,95 g/L; 7,52 ± 0,64 g/L dan 7,84 ± 1,42 g/L berturut-turut pada lama pencahayaan 20:4; 12:12 dan 20:4 jam terang:gelap. Fikoeritrin yang dihasilkan berturut-turut 0,003 mg/mL, 0,023 mg/mL, dan 0,013 mg/mL.
Porphyridium cruentum is a red-cell microalga (Rhodophyceae) with the pigment phycoerythrin content that lives in a salty water environment. The cultivation of P.cruentum can be influenced by various factors, one of which is light. The light used can be modified in terms of the duration of illumination during cultivation. This research aims to determine the growth curve of P. cruentum, the content of polysaccharides, and phycoerythrin pigments based on different photoperiods. This research consisted of three stages: 1) Cultivation and harvesting of P.cruentum with different photoperiod treatments; 2) Precipitation of polysaccharide content; 3) Extraction and concentration analysis of phycoerythrin pigment. The growth phase of P.cruentum does not show a lag phase. The polysaccharide produced for photoperiods of 24:0, 12:12, and 20:4 were 7,93 ± 0,95, 7,52 ± 0,64, and 7,84 ± 1,42 g/L, respectively. The phycoerythrin produced were 0,003; 0,023; and 0,013 mg/mL, respectively
