Title: Pemanfaatan Media Whey dalam Produksi Rekombinan Papain – Like Protease dari Virus SARS-CoV-2

Abstract: Papain-Like Protease (PLpro) dalam virus SARS-CoV-2 dianggap sebagai target yang layak untuk pengembangan obat untuk mengobati pasien penyakit Coronavirus 2019 (COVID-19). Metode yang umum digunakan untuk menghasilkan PLpro adalah melalui pendekatan rekombinan menggunakan sel inang Escherichia coli yang sederhana dan terjangkau. Namun demikian, hal ini tetap membutuhkan media pertumbuhan yang mahal (Luria Bertani/LB) untuk pertumbuhan sel inang. Penelitian ini bertujuan menginvestigasi kemungkinan whey dapat digunakan untuk memproduksi PLpro SARS-CoV-2 sebagai alternatif media pertumbuhan. Kombinasi medium whey dan LB menggunakan konsentrasi whey yang berbeda (0%, 25%, 50%, dan 100%). Kepadatan sel di setiap medium akan dipantau melalui spektrofotometer UV-Vis dan ekspresi PLpro akan diinduksi dengan menambahkan 1 mM IPTG serta dipelajari melalui SDS-PAGE. Data penelitian dianalisis menggunakan analisis deskriptif. Hasil penelitian menunjukan sel inang E. coli untuk memproduksi papain-like protease (PLpro) dari SARS-CoV2 dapat direkombinasi dengan media whey. Perlakuan terbaik ada pada komposisi medium 50% whey.
Papain-Like Protease (PLpro) in SARS-CoV-2 virus is considered a viable target for drug development to treat Coronavirus Disease 2019 (COVID-19) patients. The commonly used method for PLpro production is through recombinant approach using Escherichia coli as a host cell, which is simple and affordable. However, this still requires expensive growth media (Luria Bertani/LB) for host cell cultivation. This study aims to investigate the possibility of using whey as an alternative growth medium for producing PLpro from SARS-CoV-2. A combination of whey and LB media using different concentrations of whey (0%, 25%, 50%, and 100%) was tested. Cell density in each medium was monitored using a UV-Vis spectrophotometer and PLpro expression was induced by adding 1 mM IPTG, and analyzed using SDS-PAGE. Data were analyzed using descriptive analysis. The results showed that E. coli host cells for PLpro production from SARS-CoV-2 can be successfully recombinant using whey media. The best treatment was found in the medium composition of 50% whey.
