Abstract: Tanaman herbal menjadi salah satu sumber aktivitas antioksidan alami. Beberapa herbal digunakan sebagai bahan tunggal, tetapi juga sering digunakan sebagai kombinasi yang memberikan interaksi yang berbeda. Penelitian ini bertujuan menentukan formula terbaik kombinasi tiga tanaman herbal yaitu gandarusa, kapulaga dan temulawak terhadap kadar total fenolik, aktivitas antioksidan (metode DPPH dan FRAP), dan interaksi aktivitas antioksidannya. Hasil menunjukkan bahwa kadar total fenolik, aktivitas antioksidan metode FRAP tertinggi diperoleh pada ekstrak tunggal gandarusa, dan aktivitas antioksidan metode DPPH tertinggi pada kombinasi tiga tanaman dengan masing-masing rasio 33%. Secara umum, hasil menunjukkan interaksi adisi pada metode DPPH, dan pada metode FRAP terjadi interaksi antagonis dan interaksi sinergis. Hasil setelah dioptimasi mencapai nilai desirability 0,616 dengan konsentrasi rasio berbeda antara gandarusa, kapulaga, dan temulawak yang menunjukkan nilai prediksi dan nilai eksperimen kadar total fenolik dan aktivitas antioksidan (FRAP dan DPPH) tidak berbeda signifikan.
Herbal plants are a source of natural antioxidant activity. Some herbs are used as a single ingredient, but they are also often used in combination to provide different interactions. This study aims to determined the best formula for the combination of three herbal plants, namely gandarusa, cardamom and temulawak on total phenolic contents, antioxidant activity (DPPH and FRAP methods), and their antioxidant activity interactions. The results showed that the highest total phenolic content, antioxidant activity of the FRAP method was obtained in the single extract of gandarusa, and the highest antioxidant activity of the DPPH method was found in the combination of three plants with a ratio of 33% each. In general, the results show additive interactions in the DPPH method, and in the FRAP method antagonistic interactions and synergistic interactions occur. The results after being optimized reached a desirability value of 0.616 with different concentration ratios between gandarusa, cardamom, and temulawak which showed that the predictive value and experimental value of total phenolic content and antioxidant activity (FRAP and DPPH) were not significantly different.
