dc.description.abstractThis research aims to study and find the best concentrations of soybean extract in Tris buffer (TS)for Eta wah Cross Breed (ECB) semen diluent. Three sexually mature and healthy ECB buck were used as a source of semen . The semen was collected using an artificial vagina and evaluate macro and microscopically . Ejaculate having > 70% spem motility, > 2000x1 06mt 1sperm concentration and < 15% sperm abnormally were individually processed. Each ejaculate divided intofive tubes. Four tube was mixed with 2.5%TS (TS2.5) , 3. 75%TS (TS3.75), 5%TS (TS5), and 6.25%TS (TS6.25), one other tubes was diluted with Tris egg yolk diluent (TEY) with the concentration of spermatozoa was 50x106mt 1• The semen was stored at 5°C and observed to sperm motility and viability every 12 hours until 72 hours of storage. The result showed that TEY was the best (p<0.05) on the sperm motility as well as sperm viability compare to TS in all concentration. TS2.5 (58.33 ± 2.50%) and TS3.75 (50.56 ± 4.46%) was the best TS concentration compare to TS5 or TS6.24. It was concluded that semen diluted in TEY, TS2.5 and TS2.5 can be usefor artificial insemination purpose until 72 hour of storage.id
