dc.description.abstractThe objectives of this study were to identify polymorphisms in ovine K-casein gene exon 4 and its association with milk yield, percentage of protein and milk fat in sheep under extcnsif management at JASTRU (Jonggol Animal Studies Teaching and Research Unit) farm, Fact. of Anim Sci, Bogor Agric. University. The 83 blood sample from ewes which have record of mill\ production were used to determined polymorphism by using PCR-SSCP (Polymerase Chain Reaction-Single Strand Conformation Polymorphism) methode. The research activities were carried out through blood collection, DNA isolation, and fragment DNA amplification by PCR and separation in electrophoresis with silver staining. Electrophoretic pattern (PCR-SSCP) revealed th rce types of K-casein gene were dcsignitcd A, Band C type. The highest proportion was A type 69.57% and followed hy B 28.99% and the lowest C 1.45'Yo. The were no significant effect of types of of K-casein gene on milk production and milk quality (percentage of protein and fat) in local sheep from Jonggol.id
dc.description.abstractTujuan dari pcnclitian ini adalah mcngidcntifikasi polymorphism didalam gcn l\ascin cxon 4 dan hu-bungannya dengan susu, pcrsentase dari protein dan lemak susu pada hiri-hiri yang herada di hawah manajemcn terpadu peternakan .JASTRU (Jonggol Animal Studies Teaching and Research Unit), Fakultas Peternakan, lnstitut Pertanian Bogor. Sehanyak 83 contoh darah dari biri-biri betina yang telah mempunyai catatan memproduksi susu, digunakan untuk menentukan polymorphism dengan meng-gunal\an metode PCR-SSCP (Po~rmerasc Chain Reaction-Single Strand Conformation Po~pnorphism). Kegiatan penclitian dilaksanakan melalui lwlel.:si darah, pengisolasian DNA, amplifikasi fragment DNA dengan PCR dan pemisahan dengan electrophoresis dan pewarnaan perak. Pola electrophoretik (PCR-SSCP) didapatl•an terdiri tiga jenis gen K-kasein yaitu tipe A, B dan C. Proporsi yang paling tinggi adalah Tipe A sebcsar 69,57'1.,, diikuti oleh Tipe B sebesar 28,99°/., dan yang paling rendah adalah Tipe C sehesar 1,45%. Tidak tcrdapat pengaruh yang signifikan pada gen K-kascin produksi susu dan mutu susu (persentase lemak dan protein) pada domba lokal dari .Jonggolid
