dc.description.abstractEight microsatellite loci located on chromosome 6/BTA-6 (CSN-3, BM 143 and BM 415), on chromosome 9/BTA-9 (ETH 225 and BM 4208) on chromosome 10 /BTA-10 (BP 31, BM 1237 and BM 888) were used to analyze the genetic variation of Holstein Friesian (HF) in Baturraden Dairy Cattle Breeding Centre. The research activities were carried out through blood collecting, DNA isolating by SAMBROOK et al. (1989), fragment DNA amplified by PCR (polymerase chain reaction) and the gene frequency calculated the by NEI methods (1987). The results showed that the eight microsatellites loci exhibited a total of 33 alleles, and 14 alleles out of 33 alleles supposed alleles specific for dairy cattle in BPTU Baturraden. There were five allele had been detected in locus BM 143 and BM 4208, four alleles in locus BM415,CSN3, ETH225, BM1237, BM888 and three alleles in locus BP31. The lowest Heterozygosis per locus (h) was 0.6151 for BM415 and the highest was 0.7301 for BM888. And the average heterozygosis for all loci (H) detected was 0.6768. The genotype BB on locus BM 143 and AC on locus CSN-3 had significantly affected estimated breeding value (EBV) for milk production.id
dc.description.abstractDelapan mikrosatelit yaitu: lokus CSN-3, BM 143 dan BM 415 terletak pada kromosom 6 (BTA-6), lokus ETH 225 dan BM 4208 pada kromosom 9 (BTA-9), dan lokus BP 31, BM 1237 dan BM 888 pada kromosom 10 (BTA-10) digunakan untuk mendeteksi keragaman genetik pada sapi Friesian Holstein (FH) di Balai Penelitian Ternak Unggul (BPTU) Sapi Perah Baturraden. Tahapan kegiatan meliputi pengambilan sampel darah sapi laktasi, dan ekstraksi DNA yang dimodifikasi, amplifikasi fragment DNA dengan delapan primer khusus melalui teknik PCR (polymerase chain reaction), genotiping dan identifikasi alel, menghitung tingkat heterosigositas, frekuensi genotipe dan alela berdasarkan NEI (1987). Hasil penelitian menunjukkan ada 33 alel yang terdeteksi pada kedelapan lokus tersebut. Lima alel pada BM 143 dan BM 4208. Empat alel pada BM415, CSN3, ETH225, BM1237 dan BM888. Tiga alel pada BP 31. Ada 14 alel dari 33 alel terdeteksi kemungkinan alel spesifik sapi perah di Baturaden. Heterozigositas per lokus (h) paling rendah 0,6151 untuk lokus BM415 dan tertinggi 0,7301 untuk lokus BM888. Rataan heterozigositas (H) untuk kedelapan lokus sebesar 0,6768. Genotipe BB pada lokus BM 143 dan genotipe AC pada lokus CSN-3 berpengaruh nyata terhadap nilai pemuliaan produksi susu.id
