dc.description.abstractKeprok Garut merupakan komoditas unggulan nasional. Karakter unggul keprok Garut belum sesuai selera konsumen khususnya pada warna buah dan jumlah biji. Teknik pemuliaan mutasi dengan iradiasi sinar gamma dapat di manfaatkan untuk meningkatkan kualitas jeruk keprok Garut tanpa biji. Penelitian ini bertujuan untuk meningkatkan keragaman melalui iradiasi sinar gamma pada kalus embriogenik jeruk keprok Garut. Kalus asal nuselus di iradiasi dengan sinar Gamma pada dosis 0, 20, 40, 60, 80 dan 100 gray. Kalus diregenerasi melalui tahapan embriogenesis somatik dan diamati pertumbuhannya. Peningkatn dosis iradiasi sinar gamma pada kalus embriogenik keprok Garut menghambat proliferasi kalus dan diperoleh dosis optimal berdasarkan LD50 berada disekitar 57,87 gray. Kalus hasil perlakuan iradiasi mempunyai kemampuan regenerasi yang beragam dengan persentase efisiensi tertinggi tahap pendewasaan pada dosis 20 dan 100 gray dan tahap perkecambahan pada dosis 20 dan 40 gray. Pertumbuhan sejumlah kecambah menghasilkan 28 tunas mutan putatif dengan keragaman 0-59%. Penyambungan secara in vitro-ex vitro tunas mutan putatip sebagai batang atas dengan Japansche Citroen sebagai batang bawah setelah 4 minggu dapat bersinergis dengan persentase hidup 75-80%.en
dc.description.abstractMandarin cv. Garut is one of local citrus which has some several superiority such as easy to peel, fresh and sweet flavour, yellowish green skin and containt 12-15 seeds per fruit, but can it not compete with citrus from other countries. Quality improvement have been the subject of citrus breeding programme. The objective of this research is to increase genetic variability of Mandarin cv. Garut through Gamma rays irradiation on embryogenic callus. Callus was irradiated at doses of 0, 20, 40, 60, 80 and 100 gray and regenerated through somatic embryogenesis. The result of radiosensitivity dose for GR50 analyzed by Curve Expert 1.4 software was 58,36 gray. Observation on the growth of callus showed variation on morphology and weight of callus. At doses 0-40 gray callus growth was not inhibited, but at doses 60-100 gray callus growth was inhibited. Gamma irradiation also affected the formations of somatic embryos. After six weeks on maturation medium produced the highest percentage of efficiency at dose of 20 and 100 gray and on germination medium at dose of 20 and 40 gray. After four times subcultures in medium without plant regulator, it was produced 28 putative mutan shoots with morphological variability 0-59%. After four week grafting (in vitro and ex vitro) between putative mutan shoots as scion and Japansche Citroen as rootstock it was obtainted the growth percentage 63-75%.en
