In-vitro germination needs some treatments that result in rapid, uniform and high percentage of germination. Pummelo (Citrus maxima (Burm.) Merr.) has mucilage seed that will affect the culture. The aim of this research is to find an effective method to sterilize and to germinate Pummelo seed. The result showed that the best procedure is by peeling the outer seedcoat, 20-min disinfested in 20% bleach (5,25% w/v NaClO), two days presoak in sterile aquadest at 5oC. After that the inner seedcoat was peeled and 20-min disinfested in 20% bleach. The seeds were sowed horizontally on the top of the medium. The medium used to culture the seeds is 30% sucrosa and 2 g L-1 fertilizer (20-20-20) solidified with 7 g L-1 agar. The seeds germinated 100% at the third days after sowing.en
