An experiment was conducted to study the integrity of ram sperm plasma membrane using hypoosmotic solution as part of semen quality evaluation. Semen from six rams were collected using artificial vagina and evalusted macro- and nricroscopically. The collected semen were diluted in egg yolk-citrate extender and stored in refrigerator for 7 consecutive days. The preserved semen were examined to evaluate the motility and its plasma membrane integrity after exposed to incubation temperature for 15, 30, and 45 minutes. The result of the experiment showed a significant correlation between motility and sperm plasma membran integrity. Correlation coefficient between motility and sperm plasma membrane integrity for 15, 30, and 45 minutes incubation time were 0.96, 0.97 and 0.98 respectivelly, (P<0.01). The highest correlation (-1) was documented for 45 minutes and indicates that HOS Test should be took place at 45 minutes after incubation time. In conclusion, the sperm membrane plasma integrity could be evaluate in conjuction to motility in order to screen any cellular damage due to preservation process. The HOS Test for ram semen give the best result after 45 minute incubation time.
