Sitotoksisitas terhadap sel Hela dan Identifikasi Taksol pada Metabolit Sekunder Kapang Endofit Taxus sumatarana
Abstract
Taxus sumatrana is a rare plant that it's stem can produce taxol, an anticancer compound. Scarcity of the plant become the reason for using of it's endophyte fungus to produce Taxol. The purpose of this research is to test the cytotoxicity againts HeLa cells of Secondary Metabolites of Taxus sumatrana’s Endophytic fungus and to identify the presence of Taxol. A total of 9 isolates of endophytic fungi were isolated from the stems of the plant Taxus sumatrana. After going through the process of fermentation in PDB medium for 3 weeks, a mixture of dichloromethane extract liquid medium (PDB) and 80 % ethanol extract of biomass, tested cytotoxic activity and to identify the presence of Taxol compounds in the extract mixture. Extract mixture with fungi label 3, 5, and 7 have cytotoxic activity in BSLT test with LC50 values 13:03 ppm 3:08 ppm and 74.88 ppm. Extract fungi label 3, 5, and 7 shown to inhibit the growth of cervical cancer cells (HeLa) in MTT test with inhibition values in the range of 50 %. At LC-MS test, no Taxol compound detected
