The objective of this study was to analyze the knowledge of adolescent girls on first 1000 days of life in rural and urban areas. Design used for this study was a cross-sectional. This study was conducted in Sumenep District. Subject were 55 and 45 female high school students in urban and rural areas respectively. The knowledge of adolescent girls on first 1000 days of life was divided into four component consist of pregnancy, the care of infant 0—6 months, the care of infant 7—24 months and clean and healthy lifestyle behavior. The result showed the knowledge of adolescent girls on first 1000 days of life was low (score is<60.0%). The average score of knowledge on first 1000 days of life in urban areas (33) is higher than in rural areas (23). There is significantly differences in adolescent girls’ score between urban and rural areas (p<0.05). Knowledge of adolescent girls on pregnancy and the care of infant 0—6 months have the lower compare the other component. Participation in extracurricular activities affect the knowledge of adolescent girls on first 1000 days of life (p<0.05).
