      Uji Potensi Transmisi Vaksin Gp25 pada Bakteri Flora Normal Media Budidaya Ikan Mas Cyprinus Carpio Secara In Vitro

      Abstract
      Koi herpesvirus is a virus thats infects common carp and koi. KHV attacked almost all stadia of common carp and koi and cause mortality 80-95% of population. One way prevent the spread of KHV is with use a DNA vaccine. This research was conducted to test potential transmission of DNA vaccine encoding glycoprotein 25 toward normal flora bacteria on media cultivation of common carp Cyprinus carpio. Origin bacteria of water from common carp pond are isolated and tested the sensitivity of antibiotics ampicillin. research by adding bacteria sensitive of ampicillin with 100 μl of 12,5 μg/100 μl vaccine GP25 and incubated at temperatures 28º C for 30, 60, 180 and 300 minutes then plated on media containing antibiotic ampicillin. The growth of bacteria occurring tested existence of plasmid containing a marker resistant to ampisilin in the bacteria cell use a method of selection bacteria colonies ( cracking cells ). The results of this research show that there are no bacteria containing plasmid GP25 by cracking bacteria, electrophoresis and PCR method.
       
      Koi herpesvirus (KHV) merupakan virus yang menginfeksi ikan mas dan koi. KHV menyerang hampir semua stadia ikan mas dan koi dan menyebabkan kematian 80-95% populasi. Salah satu cara penanggulangan penyebaran KHV adalah dengan penggunaan vaksin DNA. Penelitian ini dilakukan untuk menguji potensi transmisis vaksin DNA Glikoprotein 25 (GP25) pada bakteri flora normal pada media budidaya ikan mas Cyprinus carpio. Bakteri asal kolam budidaya ikan mas diisolasi dan diuji sensitivitasnya terhadap antibiotik ampisilin. Penelitian dengan menambahkan bakteri sensitif ampisilin dengan vaksin GP25 sebanyak 100 μl dengan dosis 12,5 μg/100 μl dan diinkubasi pada suhu 28º C selama 30, 60, 180, dan 300 menit kemudian disebar pada media mengandung antibiotik ampisilin. Pertumbuhan bakteri yang terjadi diuji keberadaan gen pembawa plasmid yang mengandung penanda resisten terhadap ampisilin dalam tubuh bakteri menggunakan metode seleksi koloni bakteri ( Cracking sel). Hasil penelitian ini menunjukan bahwa tidak ada bakteri yang mengandung gen pembawa plasmid GP25 ditandai dengan tidak terdeteksi pita gen pembawa plasmid pada metode Seleksi Koloni Bakteri (Cracking sel), elektroforesis dan PCR.
