      Efikasi Vaksin dengan Metode Infiltrasi Hiperosmotik untuk Mencegah Infeksi Bakteri Streptococcus agalactiae pada Ikan Nila

      Abstract
      This study aimed to evaluate the effectiveness of the vaccine with hyperosmotic infiltration method to prevent Streptococcus agalactiae infection in Nile tilapia O. niloticus. This study used Nile tilapia with average body weight 10.59±1.07 g and length 8.68 ± 0.6 cm. Before vaccination, fish was bath immersed in salinity treatments 0, 10, 20, and 30 ppt in density 2 ind/Liter for 5 minutes. After that, the fish was vaccinated by immersion method in density 5 ind/Liter at concentration 109 CFU/mL for 30 minutes. After vaccinated, fish was kept in aquarium-sized 60 cm x 30 cm x 35 cm in density 1 ind/4 Liter. The study consisted of 6 treatments and 3 replications using complete randomized design Challenge test performed on the 10th and the 20th day at concentration 105 CFU/mL with 0.1 mL/ind, whereas the parameter observations held on days 0, 10, 20, and 30. Parameters that would observe consist of mortality rate, relative percent survival, total erythrocytes, total leukocytes, antibody titers, clinical symptoms, and water quality. The results of this study indicated that immersion in 30 ppt salinity before vaccination showed the best results with relative percent survival 76.67% in 20 days post-vaccination
       
      Penelitian ini bertujuan untuk mengevaluasi efektivitas vaksin dengan metode infiltrasi hiperosmotik untuk mencegah infeksi bakteri Streptococcus agalactiae pada ikan nila Oreochromis niloticus. Ikan nila yang digunakan memiliki bobot rata-rata 10,59±1,07 g dan panjang rata-rata 8,68±0,6 cm. Sebelum ikan uji divaksinasi, ikan direndam dalam perlakuan salinitas 0, 10, 20, dan 30 ppt dengan padat tebar 2 ekor/Liter selama 5 menit. Setelah itu, ikan divaksinasi melalui metode perendaman dengan dosis 109 CFU/mL padat tebar 5 ekor/Liter selama 30 menit. Ikan yang telah divaksinasi kemudian dipelihara dalam akuarium berukuran 60 cm x 30 cm x 35 cm dengan padat tebar 1 ekor/4L. Penelitian ini terdiri dari 6 perlakuan dan 3 ulangan menggunakan rancangan acak lengkap. Uji tantang dilakukan pada hari ke-10 dan ke-20 dengan dosis 105 CFU/mL sebanyak 0,1 mL/ekor, sedangkan pengamatan parameter dilakukan pada hari ke-0, ke-10, ke-20, dan ke-30. Parameter yang diamati meliputi mortalitas tingkat kelangsungan hidup relatif, total eritrosit, total leukosit, titer antibodi, gejala klinis, dan kualitas air. Hasil penelitian ini menunjukkan bahwa perendaman dalam salinitas 30 ppt sebelum vaksinasi memberikan hasil yang terbaik dengan tingkat kelangsungan hidup relatif mencapai 76,67% selama jangka waktu 20 hari pasca vaksinasi.
