Cervical cancer is the most common death cause of cancer in Indonesian women. One of the efforts of cancer treatment is the utilization of natural materials compounds. Some plants have potential as anticancer, one of them is bark of Pinus merkusii. The purpose of this study was to determine the cytotoxic activity of Pinus merkusii bark extract against HeLa cells by microculture tetrazolium technique (MTT) assay. The study also tested the phytochemical content and the initial screening of cytotoxicity with Artemia salina. The Highest yield of 70% ethanol P. merkusii bark extract was 6.30%. Pinus merkusii contains flavonoids, triterpenoids, saponins and tannins. The results of cytotoxicity assay using A. salina showed that the Pinus merkusii bark extract has activity and potential as anticancer indicated by its LC50 values < 1000 μg/mL. The 70% ethanol extract of P. merkusii bark has highest cytotoxic activity against HeLa cells with IC50 value of 266.01 μg/mL which was higher than methanol, water, and aceton extract with IC50 values for respectively 408.13 μg/mL, 433.51 μg/mL, dan 880.24 μg/mL.
