      Abstract
      Diabetes mellitus is a disease that cause highest mortality in Indonesia. Indonesia is the fourth biggest country with diabetes patient in world after United States, China, and India. Many research have done to find antidiabetic because diabetes patient increase quickly. Herbal antidiabetic are developed to increase patient health quality. Graptophyllum pictum leaves have bioactive compounds which potential as an antidiabetic using in vitro test. The aim of this research was to know the potency of etanolic extract of Graptophyllum pictum leaves as antidiabetic using in vivo test. This research used forty two Sprague-Dawley male rats. Those were divided into seven groups. Aloksan was given to disrupt pancreas and increase lipid peroxide concentration in rat liver. Etanolic 70% extract with four different doses were used in this research such as 25mg/kgBW (body weight), 50 mg/kgBW, 100 mg/kgBW, and 200 mg/kgBW. Extracts could repair rat pancreas. It can improve insulin secretion and reduce lipid peroxide concentration in rat liver. The experiment carried out analysis of histopathology, imunohistochemistry pancreas, and lipid peroxide concentration in rat liver. Results of pancreas analysis show the best dose to repair pancreas is 50 mg/kgBW and increase 100 % insulin production. Liver analysis show the best dose to reduce lipid peroxide concentration until 14.9% is 200 mg /kgBW. All dose can reduce lipid peroxide concentration but unsignificant (p>0.05).
       
      Penyakit diabetes mellitus menjadi salah satu penyebab kematian tertinggi di Indonesia. Jumlah pasien diabetes di Indonesia menempati urutan keempat terbanyak di dunia setelah Amerika Serikat, China, dan India. Banyak penelitian yang telah dilakukan untuk menemukan antidiabetes. Antidiabetes berbasis herbal dikembangkan untuk meningkatkan kualitas hidup penderita diabetes. Daun wungu (Graptophyllum pictum (L.) Griff) dilaporkan berpotensi sebagai antidiabetes secara in vitro. Penelitian ini bertujuan menguji khasiat ekstrak etanol daun wungu sebagai antidiabetes secara in vivo. Sebanyak 42 ekor tikus Sprague-Dawley jantan yang dibagi menjadi tujuh kelompok. Aloksan diberikan untuk merusak pankreas dan meningkatkan kadar lipid peroksida hati tikus. Ekstrak yang digunakan adalah ekstrak etanol 70% dengan empat dosis berbeda yaitu 25 mg/kgBB, 50 mg/kgBB, 100 mg/kgBB, 200 mg/kgBB. Ekstrak daun wungu mampu memperbaiki pankreas tikus diabetes sehingga meningkatkan sekresi insulin dan menurunkan kadar lipid peroksida hati. Pengujian yang dilakukan meliputi histopatologi dan imunohistokimia pankreas serta lipid peroksida hati. Hasil pengujian patologi pankreas menunjukkan bahwa dosis 50 mg/kgBB merupakan dosis terbaik yang mampu memperbaiki pankreas dan meningkatkan sekresi insulin sebesar 100 %. Dosis 200 mg/kgBB merupakan dosis yang mampu menurunkan kadar lipid peroksida hingga 14.9%. Semua dosis mampu menurunkan kadar lipid peroksida tetapi belum memberikan pengaruh yang signifikan pada tingkat kepercayaan 95% dan taraf α = 0.05.
