Karakterisasi aktivitas dialisat enzim protease fibrinolitik dari cacing tanah (Eisenia foetida) galur lokal
Abstract
Commercially available thrombolytic drugs were usually administered intraveously and priced above consumer resources. Earthworm, in several Eastern Asia countries, was known for its thrombolytic properties. Its herbal drug had been used to increase blood flow and to treat leakage in blood vessel. Thrombolytical properties of fibrinolytic protease (lumbrokinase) enzyme from local strain Eisenia foetida had never been reported. This research was purposed to characterize its enzyme activity and its specificity to fibrin as substrate. Enzyme was purified through precipitation, dialysis, and column chromatography fractionation. The highest protease activity was obtained when the dialysate sample was incubated for 10 minutes in pH 8 at 60°C which activity was 0.239 Um . Spectrophotometry based analysis showed that lumbrokinase was able to degrade fibrin up to 30.9% compared to sample without enzyme addition. Working condition to obtain fibrinolytic activity was 10 minutes incubation in pH 8 at 60°C. Lumbrokinase samples electrophoresis at each step have molecular weight between 25.200 and 29.318 kD. The highest protein concentration was found in dialysate with 1.74 m m . Obtained dialysate has protein purity up to 72.28% higher than crude extract.
