Seleksi menggunakan PCR berdasarkan marka gen badh2 pada pembentukan BC2F1 Ciherang/Mentik Wangi dan BC3F1 Ciherang/Pandan Wangi
Abstract
Commercial value and market demand (national and international) of aromatic rice is increasing. However, its less agronomy quality traits. This research is part of the development of new nontransgenic aromatic varieties with good agronomy character as nonaromatic rice through site-directed crossing. BC1F1 Ciherang/Mentik Wangi (CM) and BC2F1 Ciherang/Pandan Wangi (CP) obtained from previous studies were backcrossed with Ciherang to obtain progenies BC2F1 CM and BC3F1 CP. Selection were carried using Bradbury marker assisted PCR for CM and RM223 assisted PCR for CP. DNA samples were isolated from young leaves, PCR amplificated, and analyze using agarose 1.5% for CM and 3% for CP. Results showed heterozygous bands for both BC2F1 CM and BC3F1 CP, indicated succesful transfer of aroma badh2 heterozygous gene from Mentik Wangi or Pandan Wangi to Ciherang. DNA amplification result of BC2F1 CM using Bradbury marker obtained size of band about 580 bp, 355 bp, and 257 bp. Furthermore, RM223 marker can differentiate both the patterns of aromatic and nonaromatic rice by the band size about 160 bp and 140 bp.
