Aktivitas hepatoprotektor ekstrak metanol daun kari (Murraya koenigii .L.) pada tikus putih Sprague dawley
Activity of Curry Leaf (Muraya koenigii. L.) Methanol Extract as Hepatoprotector in Sprague dawley Rats
Abstract
Curry leaf is being thought as hepatoprotector againts liver damage caused by hepatotoxic agent such as paracetamol. This study was assesing the activity of curry leaf extract in reducing the activity of ALT (alanine aminotransferase) and AST (aspartate aminotransferase) in rats were given high dose paracetamol. Total of 25 rats were used in this research which were divided into 5 experimental groups as follows: normal control group (N), negative control group (KN) (paracetamol dose 500 mg/Kg BW were given 21 days), positive control group (KP) (curliv dose 42.86 mg/Kg BW were given 21 days), dose 200 mg/Kg BW extract group (ED200), and dose 300 mg/Kg BW extract group (ED300). Extracts were given 7 days before paracetamol treatment and continous until end of experiment. Pytochemistry test results showed that curry leaf methanol extract contain alcaloids, tannins, saponins, and steroids. Extract group had not significantly reduced ALT and AST activity (p>0.05) compared to N group. Decreasing ALT and AST activity in ED200 group is 16.21% and 33.82%. Decreasing ALT and AST activity in ED300 group is 33.82% dan 62.60%. Methanol extract curry leaf had hepatoprotector activity although not significantly differences. Daun kari diduga memiliki aktivitas hepatoprotektor terhadap kerusakan hati yang disebabkan oleh agen hepatotoksik. Penelitian ini menguji ekstrak daun kari terhadap peningkatan aktivitas alanin aminotransferase (ALT) dan aspartat aminotransferase (AST) serum darah tikus yang dicekok parasetamol dosis tinggi. Sebanyak 25 ekor tikus jantan terbagi atas 5 kelompok sebagai berikut: kelompok kontrol normal (N), kelompok kontrol negatif (KN) (parasetamol dosis 500 mg/Kg BB diberikan selama 21 hari), kelompok kontrol positif (KP) (curliv dosis 42.86 mg/Kg BB diberikan selama 21 hari), kelompok ekstrak dosis 200 mg/Kg BB (ED200), dan kelompok ekstrak dosis 300 mg/Kg BB (ED300). Pemberian ekstrak dimulai 7 hari sebelum pemberian parasetamol dan seterusnya hingga akhir percobaan. Hasil uji fitokimia menunjukkan bahwa daun kari mengandung senyawa alkaloid, tanin, saponin, dan steroid. Aktivitas ALT dan AST pada kelompok ekstrak menurun tidak signifikan (p>0.05) dibandingkan kelompok N. Penurunan aktivitas ALT kelompok ED200 sebesar 16.21% dan AST sebesar 33.82%. Penurunan aktivitas ALT kelompok ED300 sebesar 33.82% dan AST sebesar 62.60%. Ekstrak metanol daun kari dosis 200 mg/Kg BB dan 300 mg/Kg BB memiliki efek hepatoprotektor walaupun tidak berbeda nyata.
