Plant Regeneration of Pepper by Tissue Culture
Abstract
Tissue culture in papper (Piper nigrum) can be used in multiplication of new or introduced varieties and to provide i n vitro culture for other research such as protoplast culture. A series culture of later a l buds and leaf tissues. The first experiment was carry out to induce shoot growth and multiplication derived from lateral buds. The explant was planleton basal media (NS, MS 1 / 2 and MS 1/41 which was respectively enriched with the combination of BaP ( 0 . 1 ; 0.5; 1.0; 3.0 and 5.0 mg/lJ with IDA (0.1 and 1.0 f n g / l ). The second experiment was carry out to induce root growth by applying IBA (0.0; 0.1; 0.5; and 1.0 mq/l) and IAA (0.1 and 1.0 111(1/1). The third experiment tried to produce planlet from leaf tissues. The tissue was cultured in the media enriched with 2 . 4 D at the concentration of 1.0; 2.0; 4.0 and 0.0 h g / l and BAP ' of 0 . 0 ; 1.0; 5.0 and 10.0 m g / l . Resul t showed thht MS or MS 1/2 as basal media did not affect t h e number of shoots produced. The use 02 BA of 20.0 111g/l tended to in hibit the growth of multiple shoot and leaves of the culture. The application different auxine2 at different concentrations would affect the growth of roots.
