Karakter Metabolisme Etanol pada Monascus Sp.MM yang Bersifat Tahan terhadap Etanol Konsentrasi Tinggi
Abstract
A study was carried out to know ethanol metabolism character of Monascus sp. MM, which is tolerant to ethanol at high concentration. Previously inoculum of the fungus were prepared as disk agar after grown on tap water agar medium for two weeks. Each inoculum was then inoculated into media containing 40% of ethanol as sole carbon source, or 20 % of glucose and amonium (1 gll (NH4)2P04 ) or 20 % of glucose and nitrate (1 gll KN03). The cultures were incubated at 30°C. After 0, 1, 5, 6, and 8 days of incubation, supernatants were harvested for analyzing ethanol and acetic acid contents by using gas chromatography. The weight measurement of fungal mass was done by means of weighing of dry weight of the fungal mass. The dry weight was achieved by drying the fungal mass at 105 "C for 3 hours. The results showed that the fungus could grow well in 40% of ethanol medium as increase of 0.02 g the fungal mass after eight days of incubation, and ethanol concentration decreased to 22%. In this culture, acetic acid was not detected. After 3 days of incubation, 1.02 % of ethanol was detected in medium containing glucose and amonium. Ethanol (0.16 %) could be detected in medium containing glucose and nitrate at 5 days of incubation. The data indicated that the Monascus sp. MM had a capabilty of degrading ethanol and growing ai high concentration of ethanol. Therefore, this strain may be useful for analyzing defense mechanism against ethanol toxicity.
