      Abstract

Cancer has become one of the leading causes of death worldwide, including in Indonesia. Curcuma aeruginosa (temu ireng) is a medicinal plant with potential to be developed as an anticancer agent due to its ability to induce apoptosis. Fermentation using Aspergillus oryzae is expected to enhance the bioactive compound content of C. aeruginosa extract. This study aimed to evaluate the apoptosis-inducing activity of fermented and non-fermented C. aeruginosa extracts. Yeast cells were used as a model for mammalian cells. A 4% glucose solution was used as a positive control, 1% DMSO as a negative control, and 2% methanol as a solvent control. Apoptosis induction was assessed based on petite cell frequency and mitochondrial activity post-induction. The highest petite cell frequencies were observed at a concentration of 12.5 ppm for both fermented and non-fermented extracts, at 45% and 67.07%, respectively. Mitochondrial activity tested using confocal microscopy and rhodamine B staining showed a decrease in red fluorescence intensity in line with increasing concentrations of C. aeruginosa extract.
