      Rancangan Simplex Lattice Kombinasi Ekstrak Sirih Merah (Piper crocatum) dan Propolis sebagai Bahan Aktif Anti-Aging Kulit

      Abstract
      Penuaan pada kulit disebabkan oleh tingginya paparan sinar UV yang menyebabkan kadar ROS pada kulit meningkat dan degradasi kolagen akibat enzim kolagenase sehingga terjadi photoaging. Penelitian ini bertujuan menentukan kombinasi terbaik dari bahan alam sirih merah dan propolis sebagai anti-aging kulit dengan mengidentifikasi kandungan total fenolik dan aktivitas antioksidan dari kedua bahan tersebut. Selain itu, penelitian ini bertujuan juga untuk menguji aktivitas inhibisi enzim kolagenase pada kombinasi terbaik menggunakan Simplex-lattice Design (SLD). Metode pengujian yang digunakan adalah kolorimetri fenolik dengan folin-ciocalteu, antioksidan dengan DPPH, dan kolagenase dengan kit ab139443. Metode kombinasi dan optimasi menggunakan SLD. Hasil yang diperoleh dari penelitian ini adalah pada kombinasi 2, yaitu (75% propolis serta 25% sirih merah) menjadi kombinasi optimum dengan kadar total fenolik 53,94 mg GAE/gram ekstrak dan kapasitas antioksidan 9,04 mg AAE/gram ekstrak. Optimasi kombinasi mendapatkan nilai desirability 1 dari model kuartik kedua respon. Kombinasi optimum dengan konsentrasi 10 ppm memiliki inhibitor persen aktivitas kolagenase tersisa sebesar 6%.
       
      Aging of the skin is caused by high exposure to UV light which causes ROS levels in the skin to increase and collagen degradation due to the collagenase enzyme resulting in photoaging. This study aims to determine the best combination of natural ingredients red betel and propolis as anti aging skin by identifying the total phenolic content and antioxidant activity of the two ingredients. In addition, this study also aims to test the inhibitory activity of collagenase enzyme in the best combination using Simplex Lattice Design (SLD). The test methods used were colorimetric phenolic with folin-ciocalteu, antioxidant with DPPH, and collagenase with ab139443 kit. Combination and optimization methods using SLD. The results obtained from this study were in combination 2, namely (75% propolis and 25% red betel) being the optimum combination with total phenolic content of 53.94 mg GAE/gram extract and antioxidant capacity of 9.04 mg AAE/gram extract. The combination optimization obtained a desirability value of 1 from the quartic model of both responses. The optimum combination with a concentration of 10 ppm has a percent inhibitor of collagenase activity remaining at 6%.
