      Optimasi pH dan Rasio Volume-Padatan Terhadap Polifenol dan Antioksidan Daun Wungu dengan Microwave Assisted Extraction

      Abstract
      Daun wungu (Graptophyllum pictum L. Griff) merupakan tanaman yang memiliki kandungan senyawa polifenol sehingga berpotensi sebagai antioksidan. Penelitian ini bertujuan menentukan rasio volume-padatan dan pH optimum pada proses ekstraksi daun wungu dengan metode Microwave Assisted Extraction (MAE) terhadap penentuan kadar total fenolik, kadar total flavonoid dan kapasitas antioksidan menggunakan Response Surface Methodology (RSM). Ekstraksi dilakukan menggunakan one factorial dengan diawali oleh rasio volume-padatan (3:1, 5:1, 10:1, 15:1), lalu pH (2, 4, 6, dan 8). Hasil menunjukkan bahwa solusi optimasi rasio volume-padatan dan pH didapat pada3 rasio 15:1 dan pH 6 dengan nilai desirability sebesar 0,940. Verifikasi solusi optimasi dilakukan secara triplo dengan melihat nilai %Residual Standard Error (RSE) pada kadar total fenolik, flavonoid, dan kapasitas antioksidan (DPPH, FRAP) dengan nilai masing-masing sebesar -6,55; 0,54; -6,12; 6,47. Response Surface Methodology (RSM) dapat mengoptimalkan rasio volume-padatan dan pH esktraksi daun wungu dengan respon kadar total fenolik, flavonoid dan kapasitas antioksidan (DPPH, FRAP).
       
      Wungu leaves (Graptophyllum pictum L. Griff) contain polyphenol compounds with potential antioxidant properties. This study aimed to determine the optimal pH and volume-to-solid ratio for the extraction of wungu leaves using Microwave Assisted Extraction (MAE), based on total phenolic content (TPC), total flavonoid content (TFC), and antioxidant capacity, utilizing Response Surface Methodology (RSM). The extraction process involved varying the volume-to-solid ratio (3:1, 5:1, 10:1, 15:1) and pH levels (2, 4, 6, 8). Results indicated that the optimal extraction conditions were a volume-to-solid ratio of 15:1 and a pH of 6, with a desirability value of 0,940. Verification of these conditions, conducted in triplicate, revealed % Residual Standard Error (RSE) values for TPC, TFC, and antioxidant capacity (DPPH, FRAP) of -6,55; 0,54; -6,12; and 6,47, respectively. Thus, RSM effectively optimized the extraction parameters for wungu leaves, enhancing TPC, TFC, and antioxidant capacity.
