      Efikasi Vaksin Inaktif Aeromonas salmonicida untuk Mencegah Penyakit Furunkulosis pada Benih Ikan Mas Cyprinus carpio

      Abstract
      Penyakit furunkulosis adalah penyakit bakterial yang disebabkan oleh bakteri Aeromonas salmonicida. Penyakit ini menyerang komoditas budidaya ikan air tawar, salah satunya ikan mas. Vaksinasi dapat dijadikan salah satu solusi untuk mencegah penyebaran penyakit furunkulosis. Tujuan penelitian ini menguji efikasi vaksin inaktif Aeromonas salmonicida dengan dosis berbeda untuk mencegah penyakit furunkulosis dengan menganalisis respons imun spesifik dan non-spesifik pada benih ikan mas. Ikan uji yang digunakan dalam penelitian ini yaitu ikan mas dengan kisaran bobot 7,9±0,97 g dengan panjang 8,1±0,51 cm. Rancangan penelitian ini menggunakan rancangan acak lengkap dengan lima perlakuan dan tiga ulangan. Dosis vaksin yang digunakan yaitu V8 (108 CFU/mL), V7 (107 CFU/mL), dan V6 (106 CFU/mL). Parameter yang diamati yaitu total eritrosit, kadar hemoglobin, kadar hematokrit, total leukosit, aktivitas fagositik, dan titer antibodi. Vaksinasi dengan vaksin inaktif terbukti mampu meningkatkan respons imun non-spesifik dan spesifik pada ikan mas. Total eritrosit tertinggi pada perlakuan V8 sebesar 1,40±0,04(×106 sel/mm3). Kadar hemoglobin tertinggi pada perlakuan V8 sebesar 8,87±0,31 g%. Kadar hematokrit tertinggi pada perlakuan V8 sebesar 25,30±1,10%. Total leukosit tertinggi pada perlakuan V8 sebesar 5,20±0,30 (×104 sel/mm3). Aktivitas fagositik tertinggi pada perlakuan V8 sebesar 52,67±1,15%. Titer antibodi tertinggi diperoleh perlakuan V8. Dosis vaksin terbaik yaitu vaksin inaktif dengan dosis 108 CFU/mL dengan nilai RPS 71,79%.
       
      Furunculosis is a bacterial disease caused by the Aeromonas salmonicida bacteria. This disease attacks freshwater fish cultivation commodities, one of which is carp. Vaccination can be used as a solution to prevent the spread of furunculosis. The aim of this study was to test the efficacy of Aeromonas salmonicida inactivated vaccine with different doses to prevent furunculosis by analyzing the specific and non-specific immune responses of common carp. The test fish used in this study were carp with a weight range of 7,9 ± 0,97 g and a length of 8,1 ± 0,51 cm. The research design used a completely randomized design with five treatments and three replications. The vaccine dose is V8 (108 CFU/mL), V7 (107 CFU/mL), and V6 (106 CFU/mL). Parameters observed were total erythrocytes, hemoglobin levels, hematocrit levels, total leukocytes, phagocytic activity, and titer antibody. Vaccination with inactivated vaccines has been shown to increase non-specific and specific immune responses in common carp. The highest total erythrocyte was in the V8 treatment of 1,40 ± 0,04 (×106 cells/mm3). The highest hemoglobin level was in the V8 treatment of 8,87 ± 0.31 g%. The highest hematocrit level was in the V8 treatment of 25,30 ± 1,10%. The highest total leukocyte was in the V8 treatment of 5,20 ± 0,30 (×104 cells/mm3). The highest phagocytic activity was in the V8 treatment of 52,67 ± 1,15%. The highest titer antibody was obtained by the V8 treatment. The best vaccine dose is an inactivated vaccine with a dose of 108 CFU/mL with an RPS value of 71,79%.
