      Isolasi, Penapisan, dan Uji Aktivitas Antibakteri Ekstrak Fungi Laut Terhadap Bakteri Patogen Escherichia coli

      Abstract
      Colibacillosis merupakan penyakit infeksi pada ruminansia dan unggas yang disebabkan oleh bakteri Escherichia coli. Fungi laut memiliki senyawa bioaktif yang mempunyai kemampuan antibakteri. Tujuan penelitian ini adalah melakukan isolasi, penapisan, dan mengkaji aktivitas antibakteri ekstrak fungi laut yang berasal dari rumput laut dan mangrove untuk menghambat pertumbuhan bakteri patogen Escherichia coli. Penelitian dilakukan dengan beberapa tahapan yaitu isolasi, purifikasi, seleksi fungi laut, karakterisasi fungi laut, kultivasi fungi laut, ekstraksi senyawa aktif, pengujian aktivitas antibakteri, identifikasi senyawa bioaktif dengan kromatografi lapis tipis, dan uji bioautografi. Ekstraksi dilakukan dengan metode maserasi menggunakan pelarut etil asetat dan dipekatkan dengan rotary evaporator. Hasil penelitian menunjukkan bahwa terdapat 21 isolat fungi laut yang diperoleh dari rumput laut dan mangrove. Ekstrak isolat fungi laut terseleksi yaitu isolat WB 1-2 mempunyai aktivitas antibakteri terhadap E. coli pada konsentrasi 0,5; 1 dan 2 mg. Kesimpulan penelitian ini adalah ekstrak isolat WB 1-2 membentuk zona hambat (inhibisi) tertinggi pada konsentrasi 2 mg/µL sebesar 17,60±5,35 mm. Hasil uji kromatografi lapis tipis (KLT) ekstrak isolat fungi laut WB 1-2 menunjukkan adanya 5 spot reteardation factor (Rf) dengan tiga komponen warna yaitu cokelat, kuning, dan ungu. Bioautografi menghasilkan 2 spot zona hambat yaitu 18,08 dan 17,02 mm dengan nilai Rf = 0,95 dan 0,93 yang menunjukkan senyawa fenol dan flavonoid. Senyawa yang terdeteksi pada ekstrak fungi adalah senyawa golongan fenol, steroid dan flavonoid.
       
      Colibacillosis is one of the infectious diseases of ruminants and poultry caused by Escherichia coli bacteria. Marine fungi have bioactive antibacterial compounds. The aim of this research was isolate, screen, and assess the antibacterial activity of marine fungi extracts derived from seaweed and mangrove to inhibit the growth of pathogenic bacteria Escherichia coli. The research was carried out with several stages of isolation, purification, and selection of marine fungi, characterisation of marine fungi, cultivation of marine fungi, extraction of active compounds, in vitro test of antibacterial activity, identification of bioactive compounds by thin layer chromatography (TLC), and bioautography test. The extraction was performed by maceration method using ethyl acetate solvent and concentrated by rotary evaporator. The results showed that there were 21 marine fungi isolates obtained from seaweed and mangrove. Extracts of selected marine fungal isolate WB 1-2 had antibacterial activity against E. coli at concentrations of 0.5; 1 and 2 mg. The conclusion of this study is that extract of marine fungal isolate WB 1-2 showed the highest inhibition zone at a 2 mg/µL concentration of 17.60±5.35 mm. The results of TLC on extracts of isolate WB 1-2 showed the existence of 5 spots reteardation factor (Rf) with three colour components namely brown, yellow, and purple. Bioautographic test yielded 2 inlet zone spots of 18.08 and 17.02 mm at Rf 0.95 and 0.93 indicating phenol and flavonoid compounds. The compounds detected in the fungal extract were phenol, steroid, and flavonoid compounds.
