      Isolasi, Penapisan dan Kajian Antibakteri Ekstrak Fungi Laut Endofit terhadap Salmonella typhimurium

      Abstract
      Upaya pengendalian penyakit infeksi bakteri dilakukan dengan pemberian antibakteri yang alami sehingga tidak berpotensi menghasilkan residu pada produk ternak. Salah satu sumber antibakteri alami asal fungi laut didapatkan dari ekstrak alga maupun daun bakau. Penelitian ini bertujuan untuk mendapatkan isolate fungi endofit prospektif yang memiliki aktivitas antibakteri. Metode yang digunakan pada penelitian ini meliputi isolasi, pemurnian, uji antagonis, kultivasi, ekstraksi, uji antibakteri, fraksinasi dengan KLT, dan uji bioautografi. Data dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa terdapat 20 isolat yang diisolasi dari rumput laut dan daun bakau. Berdasarkan hasil uji antagonis isolate fungi prospektif adalah solate WB1-2 dari daun bakau. Isolat WB1-2 yang dikultivasi pada hari ke-9 memiliki aktivitas antibakteri maksimum sebesar 8,88 mm (level 2 mg), 6,61 mm (level 1 mg), dan 4,91 mm (level 0,5 mg). Senyawa aktif yang terkandung dalam ekstrak fungi Aspergillus terreus adalah steroid, fenol, dan flavonoid. Fraksinasi dengan KLT dan bioautografi menunjukkan adanya zona hambat 17,7 mm pada nilai Rf 0,9, 0,83 (λ= 254 nm) atau 0,8 (λ= 366 nm). Kesimpulannya isolat fungi laut daun bakau yang dikultivasi selama 9 hari memiliki zona hambat terbesar 8,88 mm (level 2 mg). Senyawa aktif yang terdeteksi pada fungi meliputi steroid, fenol, dan flavonoid.
       
      Efforts to control bacterial infectious diseases carried out by giving natural antibacterial. One of natural antibacterial sources is marine fungi including algae extracts and mangrove leaves. The aim of study was analyzed the antibacterial properties of endophytic marine fungi isolates. The methods used in this study include isolation, purification, antagonist test, cultivation, extraction, antibacterial test, fractionated using TLC, and bioautography. The data obtained were analyzed descriptively. The result showed that there were 20 isolates that obtained from mangrove leaves and algae. Based on the antagonist test, the prospective fungal isolate was WB1-2 isolate from mangrove leaves. The WB1-2 isolates that cultivated on the 9 day have maximum antibacterial activity around 8.88 mm (level 2 mg), 6.61 mm (level 1 mg), and 4.91 mm (level 0.5 mg). The active compounds contained in the extract of Aspergillus terreus were steroids, phenols, and flavonoids. Fractionation with TLC and bioautography showed an inhibitory zone of WB1-2 was 17.7 mm at Rf values of 0.9, 0.83 (λ= 254 nm) or 0.8 (λ= 366 nm). In conclusion, marine fungi from mangrove leaves that cultivated for 9 days had a largest diameter of inhibition zone 8.88 mm (level 2 mg). The active compounds of fungi were steroids, phenols, and flavonoids.
