Kadar asam urat tinggi (hiperurisemia) dapat mengindikasikan berbagai
macam penyakit seperti rematik, radang sendi, dan ginjal. Kristal mononatrium
urat monohidrat yang berbentuk jarum menimbulkan reaksi peradangan dan
nyeri hebat (gout). Deteksi dini asam urat perlu dilakukan sebagai antisipasi
keparahan hiperurisemia. Deteksi asam urat yang relevan dikembangkan ialah
teknik elektrokimia, khususnya melalui voltametri. Teknik ini relatif
sederhana, cepat, sensitif, akurat, dan preparasi sampel yang mudah. Penelitian
ini menggunakan glassy carbon elektrode (GCE) sebagai elektrode kerja
termodifikasi molecularly imprinted polymer (MIP) pada permukaan elektrode
sebagai matriks pengenalan analit. MIP merupakan polimer tercetak molekul
yang dapat memfasilitasi pengenalan molekul target secara selektif. Penelitian
ini bertujuan melakukan modifikasi GCE dengan MIP polipirola dan
mengevaluasi kinerja analitiknya pada deteksi asam urat dengan teknik
voltametri.
Elektrode yang dihasilkan berupa (GCE/MIP) digunakan untuk
mengevaluasi perilaku elektrokimia asam urat 0,2 mM dan asam askorbat 4
mM dalam larutan buffer fosfat pH 7 dengan teknik differential pulse
voltammetry (DPV) pada jendela potensial 0–1,4 V vs Ag/AgCl dengan
kecepatan payar 50 mV/detik. Selain itu, Elektrode GCE tanpa modifikasi dan
GCE non imprinted polymer (GCE/NIP) sebagai pembanding. Morfologi
permukaan elektrode GCE dan GCE/MIP dikarakterisasi dengan SEM.
Keberhasilan polimerisasi MIP ditandai dengan permukaan elektrode
termodifikasi yang lebih halus dan homogen dibandingkan dengan permukaan
GCE tanpa modifikasi yang terlihat lebih kasar atau tidak homogen.
Kinerja analitik GCE/MIP dievaluasi dengan beberapa parameter, yakni
linearitas, limit deteksi, limit kuantitasi, selektivitas dan sensitivitas
pengukuran. Respon DPV menunjukkan hubungan linear antara konsentrasi
asam urat dan arus oksidasi asam urat melalui persamaan regresi y=94,929x +
0,8219 (R2=0,9908 ) dengan limit of detection (LOD) dan limit of quantitation
(LOQ) masing-masing sebesar 1 µM dan 5 µM. GCE/MIP memiliki stabilitas
dan reprodusibilitas yang baik ditandai dengan nilai persentase relative
standard deviation di bawah 5%. Performa GCE/MIP juga dievaluasi
berdasarkan kemampuannya mendeteksi asam urat pada sampel urin segar.
Sensor berbasis GCE/MIP dapat mengenali analit asam urat dengan baik meski
ada interferens dari asam askorbat. Hal ini dibuktikan dengan nilai koefisien
selektivitas GCE/MIP yang lebih besar dibandingkan GCE dan GCE/NIP.
Pengukuran kadar asam urat pada urin menggunakan GCE/MIP memberikan
hasil sebesar 4,67 mmol/L, nilai tersebut sangat dekat dengan hasil uji
laboratorium klinis. High uric acid levels (hyperuricemia) can indicate various diseases such as rheumatism, arthritis, and kidney disease. Needle-shaped crystals of monosodium urate monohydrate cause an inflammatory reaction and severe pain (gout). Early detection of uric acid needs to be done to anticipate the severity of hyperuricemia. The relevant uric acid detection techniques developed are electrochemical techniques, especially through voltammetry. This technique is relatively simple, fast, sensitive, accurate, and easy to sample prepare. This study used a glassy carbon electrode (GCE) as a molecularly imprinted polymer (MIP) modified working electrode on the electrode surface as an analyte recognition matrix. An MIP is a molecular imprinted polymer that can facilitate the selective recognition of target molecules. This study aims to modify GCE with MIP polypyrrole and evaluate its analytical performance on the detection of uric acid by the voltammetry technique. The resulting electrode (GCE/MIP) was used to evaluate the electrochemical behavior of 0.2 mM uric acid and 4 mM ascorbic acid in a pH 7 phosphate buffer solution using differential pulse voltammetry (DPV) at a potential window of 0–1.4 V vs Ag/AgCl with a scan rate of 50 mV/sec. In addition, GCE electrodes without modification and non-imprinted polymer GCE (GCE/NIP) were used as comparisons. SEM characterized the surface morphology of GCE and GCE/MIP electrodes. The success of the MIP polymerization was indicated by the surface of the modified electrode, which was smoother and more homogeneous than the surface of the unmodified GCE, which looked rougher and inhomogeneous. GCE/MIP analytical performance is evaluated by several parameters, namely linearity, detection limit, quantitation limit, selectivity, and measurement sensitivity. The DPV response shows a linear relationship between uric acid concentration and uric acid oxidation current through the regression equation y = 94.929x + 0.8219 (R2=0.9908) with a limit of detection (LOD) and limit of quantitation (LOQ) each of 1 µM and 5 µM. GCE/MIP has good stability and reproducibility, characterized by a relative standard deviation percentage value below 5%. GCE/MIP performance was also evaluated based on its ability to detect uric acid in fresh urine samples. GCE/MIP-based sensors can recognize uric acid analytes well despite interference from ascorbic acid. This is evidenced by the value of the selectivity coefficient of GCE/MIP, which is greater than that of GCE and GCE/NIP. Measurement of uric acid levels in urine using GCE/MIP gave a result of 4.67 mmol/L. This value is very close to the results of clinical laboratory tests.
