      Respon Pemberian Kolkisin Terhadap Pertumbuhan Tanaman Alfalfa (Medicago sativa L.) Pada Kultur Jaringan

      Abstract
      Tanaman Alfalfa (Medicago sativa L.) merupakan hijauan pakan ternak jenis leguminosa yang memiliki produktivitas dan kandungan protein yang tinggi. Penelitian ini bertujuan menganalisis respon pertumbuhan tanaman Alfalfa (Medicago sativa L.) dengan pemberian kolkisin. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan 5 perlakuan dan 20 ulangan. Perlakuan yang digunakan adalah P0=0 ppm, P1=0.5 ppm, P2=1 ppm, P3=1.5 ppm; dan P4=2 ppm. Data dianalisis menggunakan analisis deskriptif dan analisis varian (ANOVA) serta diuji lanjut menggunakan uji lanjut Duncan. Peubah yang diamati antara lain tinggi vertikal tanaman, panjang daun, jumlah daun, respon warna daun, kerontokan daun, kontaminasi, penyusutan media dan biomassa. Hasil analisis ragam menunjukkan parameter panjang daun tumbuh baik pada konsentrasi kolkisin 0.5 ppm dan 1 ppm. Simpulan dari penelitian ini adalah tanaman Alfalfa memberikan respon pertambahan panjang daun yang tertinggi pada pemberian kolkisin dengan konsentrasi pemberian 0.5 ppm dan 1 ppm. Kolkisin juga memberikan pengaruh terhadap perubahan warna daun yang lebih gelap. Pemberian kolkisin hingga 2 ppm tidak memberikan pengaruh terhadap pertambahan tinggi vertikal tanaman, jumlah daun, kerontokan daun, penyusutan media, biomassa dan kontaminasi.
       
      Alfalfa (Medicago sativa L.) is a forage plant that has high productivity and protein content. This study aimed to analyze the growth response of Alfalfa (Medicago sativa L.) plants with colchicine. This study used a completely randomized design (RAL) with 5 treatments and 20 replications. The treatments used were P0=0 ppm, P1=0.5 ppm, P2=1 ppm, P3=1.5 ppm, and P4=2 ppm. Data were analyzed using descriptive analysis and analysis of variance (ANOVA) and tested using Duncan's test. The variables observed were plant height, leaf length, number of leaves, leaf color response, leaf loss, contamination, media shrinkage and biomass. The results of the analysis of variance showed that the leaf length response grew well at 0.5 ppm and 1 ppm colchicine concentrations. The conclusion of this research is that the response of colchicine to Alfalfa showed optimal leaf length increase at 0,5 ppm and 1 ppm colchicine concentrations. The administration of colchicine also gave a positive trend towards increasing darker leaf color. The administration of colchicine up to 2 ppm had no effect on the increase of plant vertical height, number of leaves, leaf loss, media shrinkage, biomass and contamination.
