| dc.description.abstract | Enzim mananase dapat diaplikasikan secara luas di dunia industri khususnya industri kertas, pakan, pangan, tekstil, detergen, ataupun farmasi. Proses produksi mananase dari Bacillus subtilis dipengaruhi oleh jenis substrat, konsentrasi substrat, pH media produksi, serta suhu fermentasi. Substrat manan murni tidak direkomendasikan untuk produksi karena harganya yang terlalu mahal. Tujuan penelitian ini adalah menentukan jenis substrat alternatif terbaik dari berbagai biomassa manan untuk optimasi produksi mananase dari Bacillus subtilis yang diisolasi dari Laut Bali. Berdasarkan penelitian ini, porang merupakan substrat alternatif terbaik untuk produksi mananase dari Bacillus subtilis dengan nilai aktivitas enzim sebesar 11.29 U/mL. Aktivitas mananase optimum 37,70 U/mL diperoleh dalam 24 jam fermentasi dengan konsentrasi substrat, pH dan suhu masing-masing 1%, pH 8 dan 50oC. Kata kunci: Bacillus subtilis, manan, mananase | en |
| dc.description.abstract | Mananase can be applied widely in the industrial world, such as paper, feed, food, textiles, detergent, or pharmaceutical industries. Mannanase production process is influenced by the type of substrate, substrate concentration, pH of the media production and fermentation temperature. Mannan pure substrate is not recommended because it is too expensive. The aim of this research was to determine alternative substrates of various mannan biomass and the optimum conditions for production of mananase by Bacillus subtilis isolated from Bali sea. Based on this research, porang was the best alternative substrate for production of mananase by Bacillus subtilis with enzyme acitivities of 11.29 U/mL. The maksimum mannanase activity of 37.70 U/mL was achieved following 24 hours of fermentation when substrat concentration, pH and temperature were controlled at 1%, pH 8 and 50◦C respectively.
