Abstract: Xantorizol merupakan senyawa penciri pada temu lawak yang memiliki aktivitas antibakteri dan antijamur. Penelitian ini telah mengembangkan metode ekstraksi dan isolasi senyawa xantorizol dari temu lawak (Curcuma xanthorrhiza Roxb.). Teknik maserasi dan sokletasi dilakukan untuk mengekstraksi xantorizol dengan metanol, dietil eter, dan n-heksana sebagai pelarut. Pemurnian selanjutnya menggunakan kromatografi kolom dan kromatografi lapis tipis preparatif. Xantorizol hasil isolasi dicirikan berdasarkan spektrum ultraviolet-tampak dan inframerah juga kromatogram gas-spektrum massa serta ditentukan kemurniannya dengan kromatografi cair kinerja tinggi. Ekstrak n-heksana hasil maserasi (168 mg/g) memiliki kandungan xantorizol lebih tinggi dibanding ekstrak lainnya. Fraksi ke-4 hasil fraksionasi ekstrak n-heksana dengan kromatografi kolom memiliki rendemen akhir dugaan xantorizol sebanyak 0.016 % dengan tingkat kemurnian 87.4 %. Xanthorrhizol is a marker compound of temu lawak and has antibacterial and antifungal activities. In this study, we developed an extraction and isolation methods of xanthorrhizol from temu lawak. Maceration and soxhlet extraction were used to extract the xanthorrhizol by using methanol, diethyl ether, and n-hexane as solvents. Xanthorrhizol was futher purified using column chromatography and preparative thin-layer chromatography. The isolated xanthorrizhol was characterized based on ultraviolet-visible and infrared spectra as well as gas chromatogram-mass spectra and the purity was determined by using high performance liquid chromatography. n-Hexane extracts by maceration method contained the higher yield (168 mg/g) as compared to other extracts. The fourth fraction of n-hexane extract from fractionation using column chromatography contained 0.016 % of xanthorrhizol with 87.4 % level of purity.
