Abstract: Penggunaan fage litik sebagai biokontrol telah dilaporkan dapat menurunkan populasi bakteri resisten antibiotik. Penelitian ini bertujuan untuk mendapatkan fage litik yang mampu melisiskan Salmonella Sp. Resisten antibiotik yang diisolasi dari penderita diare. Fage litik diisolasi dari limbah cair rumah tangga, isolate fage diperbanyak dan diuji kemampuan menurunkan populasi inangnya, perpecahan sel inang diamati pada mikroskop electron SEM. Hasil isolasi dan pemurnian faga diperoleh empat isolat faga, ialah faga FR 15, FR 19, FR 38 dan FR 84. Setiap faga spesifik untuk inang masingmasing. Penambahan konsentrasi faga dari ± 10 000 PFU mL-1 dari isolat FR 38 memiliki aktivitas tertinggi. Faga ini mampu mengurangi kekeruhan populasi sel Salmonella sp. setelah 5 jam diinokulasi faga. Selain itu pada kepadatan ± 30 000 PFU mL-1 dari isolat FR 84 memiliki aktivitas melisis sel tertinggi. Faga ini mampu mengurangi sel Salmonella sp. secara signifikan dalam waktu 1 jam. Hasil penelitian ini diharapkan dapat diterapkan sebagai biokontrol air dan makanan.
Lytic phage has been reported use as biocontrol for decreasing antibiotic resistant bacteria population. The aim of this research is to get lytic phage that could lysised antibiotic resistant Salmonella sp. that isolated from patients with diarrheal disease. Lytic phage was isolated from household water waste, isolated phage was multiplied and ability for decreasing the host population was tested, observed in SEM electron microscope. There are four results of phage isolation and purification, namely FR 15, FR 19, FR 38, and FR 85. Every phage was specific for each host. Increment of phage concentration from ± 10000 PFU mL-1 of FR 38 has a highest activity. This phage could decrease Salmonella sp. cell population after 5 hour inoculation, but in ± 30 000 PFU mL-1 density FR 84 have a highest activity for cell lysising. This phage could decrease Salmonella sp. cell significantly in 1 hour. The result of this research could be expected as environmental biocontrol for Salmonella contamination
