Abstract: Tujuan penelitian ini adalah mendapatkan alternatif proses konversi lignoselulosa dari tanaman jagung menjadi etanol melalui proses Sakarifikasi dan Ko-fermentasi Simultan (SKFS). Biomassa diberi perlakuan awal dengan Ca(OH)2 dan hidrotermolisis, dilanjutkan prehidrolisis enzimatik pada suhu 50 OC selama 24 jam. Selanjutnya sakarifikasi menggunakan enzim selulase, xilanase dan β-glukosidase, sedangkan fermentasi menggunakan dua biakan campuran yaitu Saccharomyces cereviseae dan Pichia stipitis (Biakan I) serta Zymomonas mobilis dan Pichia stipitis (Biakan II). Waktu inkubasi SKFS adalah 24, 48, 72, dan 96 jam. Gula reduksi setelah prehidrolisis sebesar 11,63 g/l, sedang produksi etanol tebaik mencapai 6,31 g/l untuk biakan I dan 9,94 g/l untuk biakan II dengan waktu inkubasi yaitu 72 jam. Gula yang terkonversi pada kondisi terbaik untuk biakan I dan II adalah 32,00 % dan 53,03 %.
In this research , Simultaneous Saccharification and Co-Fermentation (SSCF) was developed to convert corn stover lignocellulosic biomass to ethanol. The corn stover biomass was pretreated using Ca(OH)2 and hydrothermolysis, then an enzimatic prehydrolysed at 50 OC, 24 h was conducted prior to SSCF. In the saccharification process, cellulase, xylanase and β-glucosidase were used, whereas for the co-fermentation two mixed cultures applied were culture I (Saccharomyces cereviseae and Pichia stipitis) and culture II (Zymomonas mobilis and Pichia stipitis). SSCF was done at incubation time of 24, 48, 72, and 96 hours. The sugar after prehydrolysis was 11,63 g/l, meanwhile the higest ethanol production of 6,31 g/l and 9,94 g/l were reached for for cultures I and culture II at 72 hours of incubation time. The sugar which converted in the process of the culture I and II were 32.00 % and 53.03 % respectively.
