Abstract: | The aim of this study was to identify the genetic polymorphisms of calpastatin (CAST), calpain 3 (CAPN3) and myostatin (MSTN) on local sheep at Jonggol Animal Science Teaching and Research Unit (JASTRU). A total number of 294 blood samples were collected from JASTRU. The identification of polymorhism in CAST and CAPN3 genes performed by using Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) while MSTN gene by using PCR-SSCP methods. The results showed that CAST| Msp I, CAST| Nco I and CAPN3| Ma eII loci were polymorphic, whereas The MSTN locus was monomorphic for G (1.0). The frequency of allele M (0.87) on the locus (CAST| Msp I) higher than the N allele (0.13). At locus CAST| Nco I, the frequency of allele M (0.96) higher than the N allele (0.04). At the CAPN3| Mae II, allele G (0.85) and allele T (0.15). Locus CAST| Nco I has higher observed heterozygosity (Ho = 0.92) compared to CAPN3| Mae II and CAST| Msp I (Ho = 0.74-0.77), however has lower compared to CAPN3| Mae II and CAST| Msp I in expected of heterozygosity (He = 0.08 vs 0.23-0.26) and in index fixation (Fis = -0.04 vs 0.03-0.12). Penelitian ini bertujuan mengidentifikasi keragaman genetik gen pengontrol kualitas daging yaitu gen calpastatin (CAST), calpain 3 (CAPN3, dan myostatin (MSTN) pada domba lokal di Unit Pendidikan dan Pelatihan Peternakan (UP3) Jonggol. Sampel darah domba dari UP3 Jonggol yang digunakan sebanyak 294 diekstraksi untuk mendapatkan sampel DNA genom. Pendeteksian keragaman pada gen CAST dan CAPN3 dilakukan menggunakan pendekatan Polymerase Chain Reaction – Restriction Fragment Length Polymorphi sm (PCR-RFLP), sedangkan pada gen MSTN dilakukan menggunakan pendekatan P o l y m e r a s e C h a i n R e a c t i o n - S i n g l e S t r a n d Comformation Polymorphism (PCR-SSCP). Hasil pendeteksian keragaman lokus CAST| Msp I dan CAPN3|MaeII menghasilkan dua alel (M dan N) dan tiga genotipe (MM, MN dan NN), sedangkan lokus CAST| Nco I menghasilkan dua ale (M dan N)l dan dua genotipe (MM dan MN). Aplikasi PCR-SSCP pada gen MSTN menemukan hanya satu alel G (1,0). Lokus CAST| Msp I, CAST| Nco I dan CAPN3| Mae II bersifat polimorfik. Frekuensi alel M (0,87) pada lokus (CAST| Msp I) lebih tinggi dari Alel N (0,13). Pada lokus CAST| Nco I, frekuensi alel M (0,96) lebih tinggi dibandingkan alel N (0,04). Pada lokus CAPN3| Mae II, alel G (0,85) dan alel T (0,15). Nilai heterozigositas pengamatan (Ho) berkisar antara 0,74 - 0,92, lebih tinggi dari nilai heterozigositas harapannya (He) yang berkisar antara 0,08–0,28. Nilai Fis domba UP3 Jonggol dalam kisaran (-0,04) - 0,12. Nilai Fis yang lebih besar ditemukan pada lokus CAPN3| Mae II (0,12). |
