Deteksi Kerusakan Morfologi Spermatozoa Parent Stock Ayam Arab Golden Red dengan Pewarnaan Eosin Nigrosin dan Carbofuchsin.

Abstract

Golden Red-arab chicken is one of local Indonesian chicken utilyzed as layer type. This study aimed to identify sperm morphological abnormalities of arab chicken using eosin nigrosin and carbofuchsin staining method. Semen was collected from Golden Red-arab chicken parent stock belong to 'Trias Farm' using massages techniques. Results demonstrated that sperm abnormalities of Golden Red-arab chicken was 30.3±11.38%, mostly in the form of bent tail. There was significantly differences among individual chickens on the number of sperm abnormalities (P<0.05). Eosin-nigrosin staining was more acurate to evaluate sperm abnormalities (P<0.05), compared to carbofuchsin. Chloramin and alcohol solution contained in carbofuchsin disturb sperm membrane, cause difficulties in sperm abnormality observation. We highly recommend to use eosin nigrosin staining method for evaluation of chicken sperm morphology.
