Abstract: The research was conducted to evaluate the effectiveness of lerak extracts and probiotics to reduce enteric methane production in vitro. The design of experiment was a randomized block design consisting of 4 treatments i.e. P1 = control (without additives); P2 = control + 1 ml liquid Saccharomyces cerevisiae isolates; P3 = control + 1 ml Lerak Extract; P4 = control + 1 ml Lerak Extract + 1 ml liquid S. cerevisiae isolates. The measured variables were gas production, dry matter digestibility (DMD), organic matter digestibility (OMD), the total gas production, methane production, NH3, protozoa population, total bacterial population, and production of volatile fatty acids (VFA). The results showed that the addition of treatment additives (P3) significantly (P<0.05) increased organic matter digestibility compared liquid treatment isolates of S. cerevisiae as well as combination treatment isolates of S. cerevisiae liquid with lerak extract. The treatment of lerak extract, liquid isolates of S. cerevisiae and their combinations had no significant effect on pH, N-NH3, total bacterial population, protozoa population, total gas production and methane production. It concluded combination 1 ml lerak extract and 1 ml liquid S. cerevisiae isolates gives the highest effect in total gas production 102.42 ml with the lowest percentage of methane 24.78% on in vitro experiments
