Situ Bagendit is one of the rice variety that sensitive to aluminum (Al) toxicity. The variety can be developed to an Al-tolerant variety because it can be planted in both dry and wet field. The effort to develop Al tolerant varieties could be done by inserting Al tolerance genes. An Al-tolerance gene, called B11 gene has been inserted to Situ Bagendit genome by in-planta technique and produced T1 generation. The research objective was to evaluate the stability of B11 gene insertion in T1 and T2 generation of rice transgenic SB-16. The evaluation was done by rice seed selection using hygromycin antibiotic with concentration 20 mg/l, molecular analysis using polymerase chain reaction (PCR) technique, and plant phenotype observation. The result of this research showed that B11 gene had been integrated in T1 plant based on molecular analysis and plant phenotype observation. The result of seed selection showed that T2 generation still segregated for B11 gene.en
