Title: Isolasi Senyawa Antitumor dari Karang Lunak Sarcophyton sp. Kepulauan Seribu

Abstract: Sarcophyton sp. is a soft corals species which can dominate the living in benthic environment. The endurance of Sarcophyton sp. in low water conditions (high eutrophication) and its dominating character have been reported due to its bioactive compound as antitumour. This study aimed to isolate the antitumour compound in Sarcophyton sp. from Panggang Island south section, Seribu Islands National Park. Brine shrimp lethality test was used as preliminary screening test and found the crude methanol extract of dominating soft coral was the most toxic extract at 100 ppm (50% lethal concentration), compared with similar species live in hard corals or abiotic environment. Fractionation of the crude extract with solid phase extraction column resulted 4 fractions. The F2 fraction was the most toxic (50% inhibition concentration, IC50 37.675 ppm). Purification of F2 with preparative high performance liquid chromatography resulted 20 subfractions. Based on purity index from photodiode array detector, there were 3 pure isolates, all with cytotoxic activity against MCF-7 cell line, namely F25 (IC50 36.04 ppm), F30 (IC50 71.030 ppm), and F37 (IC50 67.227 ppm).
