One of the importance thing of plant genetic resources is cacao (Theobroma cacao L). Seed of cacao is defined are seeds which are sensitive to desiccation, chilling and cannot be dried below a certain critical moisture level {recalcitrant}. 1t's essential to recognize that owing to the various problems and limitation encountered with both genetic reserves. Cryopreservation currently offers the only safe and cost effective option for the long-term conservation of genetic resources. Somatic embryo from zygotic embryo used as plant material because cell of somatic embryo still in globular fase. Embryo can still live and has a cal/us (13.33%) with cryoprotectant's combination contain 15% dimethysulfoxide (DMSO) and 15% glycerol, thawing 35 °C, and grow at MS medium + picloram 0.1 g Lˉ¹ + thiamin 0,2 g Lˉ¹ + IAA 1 ml Lˉ¹ + kinetin 0,1 ml Lˉ¹en
