Abstract: Kumis kucing (Orthosipon aristatus) is a medicinal plant containing sinensetin which is potential as antioxidant, antibacterial, and has diuretic activity. Because of the benefits of sinensetin, optimization for extraction of sinensetin by maceration and comparison of solvent and extraction conditions by varying material-solvent ratio, time of extraction, and number of extraction were investigated. The result showed that ethanol was the best solvent. The content of sinensetin was analyzed by high-performance liquid chromatography with the mobile phase methanol, H2O (pH 3.0), and tetrahydrofuran (45:50:5). Optimization by using regression analysis gave regression equation as: content = 5.564 + 3.265(time) - 8.223(number) - 2.017(ratio) - 0.204(time2) - 0.172(number*time) + 1.350(number2) - 0.973 (ratio*time) + 3.840(ratio*number) with R2 = 96.90%. The optimum condition of extraction was obtained at 4.72 hours of time of extraction, 2-3 times of number of extraction, and 1.12 g:100 ml of material-solvent ratio. The content of sinensetin was 5.29 mg/g. The model showed that time of extraction, material-solvent ratio, and number of extraction had a significant effect at α = 0.05 to the content of sinensetin.
