Abstract: This experiment was aimed to obtain the best combination formulations of vitamin and sucrose concentrations of the medium, and the best combination between types of cytokinins and its concentration on inducing the best multiplication of line clone Protocorm Like Bodies (PLBs) Spathoglottis plicata orchid in lagre number using Murashige and Skoog (MS) solid medium in vitro. PLBs were used as an explant materials was grown on MS modification medium that they were accordance with the treatment. This experiment was arranged in Completely Randomized Design (CRD) with two factors. The first factors was formulations of vitamin composition and concentrations of sugar medium with consists of four different formulations (J1 = vitamin MS + sucrose 30 g L-1, J2 = vitamin B5 + sucrose 30 g L-1, J3= vitamin MS + sucrose 40 g L-1, J4 = vitamin B5 + sucrose 40 g L-1). The second factors was seven level combination of cytokinin types and it concentrations (S0 = without cytokinin (controle), S1 = 20 μM BA, S2 = 40 μM BA, S3 = 20 μM kinetin, S4 = 40 μM kinetin, S5 = 75 ml L-1 coconut milk, and S6 = 150 ml L-1 coconut milk). The best growth and multiplication of PLBs orchid S. plicata produced on MS medium B5 vitamins modification combined with 30 g L-1 sucrose concentration. There were available on MS medium enrichment with 75 ml L-1 coconut milk as well as on MS medium supported by 20 μM BA based on the higest number of PLBs per explant, number of plantlets per explant, number of roots formed, plantlet height and visual appearance and performance of the observations at 6 mst.
